摘要
目的:探讨蒌芩止嗽煎能否提高青霉素在慢性阻塞性肺疾病急性加重(AECOPD)痰热证大鼠模型肺组织中的转运能力。方法:80只Wistar雄性大鼠随机均分为模型组、氨溴索组、二陈汤组、蒌芩止嗽煎组。用熏吸香烟加气管内注射内毒素及鼻腔滴入金黄色葡萄菌,鼓风干燥箱中进行风热刺激的方法建立大鼠AECOPD痰热证模型。用高效液相色谱-质谱法检测各组大鼠不同时段生物标本中青霉素的浓度。结果:15min血清及支气管肺泡灌洗液中蒌芩止嗽煎组药物浓度较模型组、氨溴索组及二陈汤组显著增高(P<0.05,P<0.01);肺组织中各组比较无统计学意义,但中位数比较显示蒌芩止嗽煎组药物浓度最高。30min血清、肺组织各组比较无统计学意义,但在灌洗液中蒌芩止嗽煎组药物浓度均低于氨溴索组、二陈汤组和模型组(P<0.05)。结论:清热化痰法指导下的蒌芩止嗽煎能提高青霉素在AECOPD痰热证大鼠模型肺组织中的药物浓度,即能增强其在肺组织中的转运能力,为临床选择中药作为COPD急性加重期抗生素治疗的辅助用药提供了实验依据。
Objective:To study whether Louqin Zhisou Decoction was capable of enhancing the delivery of penicillin in lung tissue of rat model of chronic obstructive pulmonary disease during acute exacerbation period(AECOPD) with phlegm and heat syndrome.Methods:Eighty Wister male rats were randomly divided into the model group,ambroxol group,Erchen Decoction group,Louqin Zhishou Decoction group.The rat model of AECOPD with phlegm and heat syndrome was established with multiple etiological factors including exposure to smoke,intratracheal instillation of lipopolysacchricle(LPY),intranasal instillation of staphylococcus aureus,and wind-heat stress in an air dry oven.The HPLC-MS/MS was used to detect the concentration of penicillin at different times of this study.Results:Louqin Zhishou Decoction significantly increased the concentration of penicillin in serum and bronchoalveolar lavage fluid at time point-15 min when compared with the model group,the ambrohexel group and the Erchen Decoction group(P〈0.05,P0.01).Although the results of median showed that the concentration of penicillin in the lung tissue was highest in the Louqin Zhishou Decoction group,no difference was found in the lung tissues.The concentration of penicillin in serum,and lung tissue at time point-30 min of the four groups had no statistical difference.The concentration in bronchoalveolar lavage fluid,Lougin zhishou Decoction group was lower than ambroxol group,Erchen Decoction group and model group(P0.05).Conclusion:Louqin Zhisou Decoction could increase the concentration of penicillin in rat model of AECOPD with phlegm and heat syndrome that means Louqin Zhisou Decoction is capable of enhancing the delivery of penicillin in lung tissue.This study will provide fundamental experiment basis for using Chinese herb medicine as agents to assist antibiotic treatment for AECOPD.
