摘要
目的:观察肝豆汤调控TX小鼠肝细胞内铜转运通路的分子靶点,研究其调控机制。方法:免疫荧光双标记经激光共聚焦显微镜观察由含肝豆汤兔血清孵育后TX小鼠肝细胞内ATP7b蛋白的亚细胞定位变化。结果:ATP7b蛋白在DL小鼠肝细胞内定位于反面高尔基体管网状结构(TGN),位于高铜环境中则转运至顶端膜区,去除高铜环境则重新定位于TGN;ATP7b蛋白在TX小鼠肝细胞内不能定位于TGN及顶端膜区域,去除高铜环境也不能促使ATP7b蛋白重新定位于TGN。中药肝豆汤可促使ATP7b蛋白逐步重新定位于TGN。结论:肝豆汤可以通过诱导ATP7b蛋白重新定位于TGN,进而发挥细胞内排铜作用,改善Wilson病的临床症状。
Objective:To observe the molecular target that Gandou Decoction regulates on the pathway of copper transportation in the hepatocytes of toxic milk(TX) mice and study the mechanism of regulation.Methods:By the method of immunofluorescent double-labeling,it was observed that the ATP7 b protein subcellular location in the TX mouse hepatocytes incubated in rabbit serum with Gandou Decoction.Results:The ATP7 b protein was located in the trans Golgi network(TGN)in hepatocytes of DL mice.When the hepatocytes were in the environment of high-concentration cupricion,the ATP7 b was transferred to the apical region and returned to the TGN while off that environment.The ATP7 b protein in hepatocytes of TX mice was neither located in the TGN,nor in the apical region even given the same environment as above.Gandou Decoction could stimulate the translocation of ATP7 b protein of TX mice from the cytoplasm to the TGN.Conclusion:Gandou Decoction could stimulate the translocation of ATP7 b protein to TGN in the hepatocytes of TX mice,and further to expel the cupricion,which may relieve the symptoms of Wilson disease.
