短葶山麦冬内生真菌分离鉴定及抗氧化活性 被引量:4

摘要 以福建短葶山麦冬主产区之一泉州罗溪镇采集的1 a生和盆栽6 a生短葶山麦冬为研究材料,分别从叶、须根和块根中分离内生真菌,测定根部内生真菌的抗氧化活性.结果表明:1 a生块根的内生真菌分离率高于6 a生,须根中分离的内生真菌数量最少.以总抗氧化能力为指标,评价从根部分离得到的18株菌株的抗氧化活性,对其进行降序排序,筛选前5株菌株,测定其清除DPPH自由基、羟自由基和超氧阴离子自由基的能力,结果发现,d5菌株发酵产物的清除效果最为明显.进一步测定其乙酸乙酯萃取部位3种自由基清除能力表明,d5菌株对DPPH自由基和羟自由基的能力最强,EC50分别为0.572和1.257 mg/m L.经真菌18 s r DNA序列比对测定d5菌株属于青霉菌属,可以作为进一步研究的对象. Materials were collected from Quanzhou primary production zone of Liriopemuscari( Decne.) Bailey.The endophytic fungi of leave,fibrous and tuber roots from Liriopemuscari( Decne.) Bailey of one year and six yeas were isolated,and antioxidant activity of endophytic fungi from roots were measured.The results showed that the isolation rate of tuber root of one year was higher than that of six years. The endophytic fungi number from fibrous roots was least. The total antioxidant capacity of 18 strains isolated from roots were determined,and 5strains with better antioxidant capacity were screened out,and their scavenging activity of DPPH,hydroxyl radical and superoxide radical were explored.Among them,the fermentation broth extracts of d5 exhibited best scavenging effects.Hence,the antioxidant activities of the ethyl acetate extracts of d5 was further evaluated. The capacity of scavenging the DPPH and hydroxyl radical from d5 were the strongest,and their EC50 were 0.572 mg / m L and 1.257 mg / m L,respectively.The 18 s r DNA sequence analysis suggested that d5 strains belonged to Eupenicillium sp.and were worth of making further study.
