摘要 目的:研究过氧化氢人脐静脉内皮细胞(H_2O_2)对白细胞介素(IL)-37表达的影响。方法:用实时荧光定量PCR法和Western blot法检测经系列浓度H_2O_2作用不同时间的人脐静脉内皮细胞中IL-37 mRNA和蛋白水平的变化。结果:实时荧光定量PCR结果显示:不同浓度H_2O_2作用人脐静脉内皮细胞后,IL-37 mRNA水平升高,并呈浓度依赖(P<0.01)。Western blot结果表明,H_2O_2作用人脐静脉内皮细胞20 h后,IL-37蛋白水平较对照组显著升高(P<0.01)。结论:H_2O_2能够上调人脐静脉血管内皮细胞IL-37的表达,为进一步研究IL-37在细胞氧化应激状态下的作用奠定了一定基础。 Objective:To investigate the effects of hydrogen peroxide ( H2 O2 ) on the expression of interleukin-37 ( IL-37 ) in EA. hy926 cells. Methods:The expressions of IL-37 mRNA and protein in different time-points of human umbilical vein endothelial cell treated with different concentration of H2 O2 were detected by qRT-PCR and Western blot. Results:The results of qPCR showed that the levels of IL-37 mRNA in human umbilical vein endothelial cell treated with different concentration of H2 O2 significantly increased, which was the concentration-dependent(P 〈0. 01). The results of Western blot showed that the level of IL-37 protein in human umbilical vein endothelial cell treated with H2O2 for 20 h was significantly higher than that in control group(P〈0. 01). Conclusions:The expression of IL-37 in human umbilical vein endothelial cell can be up-regulated by H2 O2 ,which may be helpful to study the role of IL-37 in cellular oxidative stress.
