导入iaaM与ipt双价基因的转基因大豆检测及表达效果研究

摘要 大豆是我国的重要的油料作物和经济作物,但其单产却远远低于欧美国家,因此提高其单产育种对国民的生产实践具有重要的意义。主要通过GUS组织化学染色分析、PCR检测、Southern blot检测技术对转iaa M与ipt双价基因的T1代转基因大豆进行检测,结果表明有5个株系iaa M和ipt基因以单拷贝的形式整合进了大豆基因组。采用半定量RT-PCR检测表明有4个株系外源双价基因高效表达。最后经产量性状考种结果表明:有3个株系的表现较好,其中株系4增产12.85%。 Soybean is an important oil and crop. However, the unit production is far lower than that in American and European countries. So it is very important to improve the unit yield breeding of the national production practice. Through GUS assay, PCR and Southern blot analysis, the T1 genetically transgenic soybean with iaa M and ipt was detected. The results indicate that 5 lines of iaa M and ipt gene was integrated into the genornic of the transgenic plants in the form of single copy. Semi-quantitative RT-PCR shows that there is a high efficient expression of exogenous binary genes in 4 lines. After measuring their yield characters, we found that 3 lines were better than others, and one line (line 4) of them increased the production to 12.85%.
