饱和氢盐水对心肌缺血再灌注损伤老年大鼠心肌细胞线粒体损伤的影响 被引量:3

摘要 目的探讨饱和氢盐水对心肌缺血再灌注损伤老年大鼠心肌细胞蛋白激酶C(PKC)/热休克蛋白90(HSP90)信号通路和线粒体损伤的影响。方法选取18月龄健康老年清洁级雄性SD大鼠150只,采用随机数字表法分为5组:空白对照组、假手术组、缺血再灌注组、饱和氢盐水组、生理盐水组,每组30只。HE染色观察心肌病理学。透射电镜观察线粒体超微结构。蛋白质印迹法和RT-PCR法检测心肌细胞PKC、HSP90、缝隙连接蛋白43(Cx43)、Cx45蛋白及PKC、HSP90、Cx43、Cx45mRNA表达。结果与空白对照组和假手术组比较,缺血再灌注组、饱和氢盐水组和生理盐水组大鼠心肌缺血再灌注后12h和24h心肌细胞线粒体损伤程度评分明显升高,PKC、HSP90、Cx43、Cx45蛋白及PKC、HSP90、Cx43、Cx45mRNA表达明显增加,差异有统计学意义(P<0.05)。与缺血再灌注组和生理盐水组比较,饱和氢盐水组大鼠心肌缺血再灌注后12h和24h心肌细胞线粒体损伤程度评分明显降低[(1.34±0.12)分vs(2.54±0.28)分、(2.61±0.29)分,(1.27±0.13)分vs(2.57±0.27)分、(2.64±0.28)分,P<0.05],PKC、HSP90、Cx43、Cx45蛋白及PKC、HSP90、Cx43、Cx45mRNA表达明显下调,差异有统计学意义(P<0.05)。结论饱和氢盐水可减轻线粒体损伤,从而减轻老年大鼠心肌再灌注损伤。 Objective To study the effect of satuated hydrogen saline on PKC/HSP90 signaling pathways and mitochondrial injury in aged rats following myocardial ischemia reperfusion injury. Methods One hundred and fifty healthy male SD rats aged 18 months were randomly divided into control group,sharn operation group, myocardial ischemia reperfusion injury group, satuated hy-drogen saline group, and normal saline group (30 in each group). Their myocardial pathology was observed with HE staining. Ultrastucture of mitochondria was detected by transmission electron microscopy. Expressions of PKC, HSP90, Cx43, and Cx45 protein and mRNA were detected by Western blot and reverse transcriptase polymerase chain reaction respectively. Results The score of mitochondrial injury and the expression level of PKC, HSP90,Cx43, Cx45 protein and mRNA in myocardial cells were significantly higher in myocardial ischemia reperfusion injury group,satu- rated hydrogen saline group and normal saline group than in control group and sham operation group, and were significantly lower in saturated hydrogen saline group than in myocardial ischemia reperfusion injury group and normal saline group at 12 and 24 h after ischemia reperfusion (1.34±0.12 vs 2.54±0.28 and 2.61±0.29;1.27±0.13 vs 2.57±0.27 and 2.64±0.28,P〈0.05). Conclusion Saturated hydrogen saline can attenuate the myocardial reperfusion injury in aged rats by reducing their mitochondrial reperfusion injury.
