摘要
目的:探讨大黄虫丸加黄芪、水红花子后对拮抗大鼠肝纤维化作用的影响。方法:将35只SD大鼠随机分为正常组、模型组、秋水仙碱组、大黄虫丸组和大黄虫丸加味组进行造模和实验共12周,于12周末采集组织样本,通过病理染色评价大鼠肝脏炎性反应及纤维化程度,通过RT-PCR技术检测基质金属蛋白酶(MMP-1)、金属蛋白酶抑制因子-1(TIMP-1)对应的m RNA表达水平。结果:与正常组相比,模型组MMP-1 m RNA降低、TIMP-1 m RNA升高(P<0.05,P<0.01);与模型组相比,大黄虫丸组、大黄虫丸加味组MMP-1 m RNA有升高趋势、TIMP-1 m RNA有降低趋势,其中大黄虫丸加味组差异显著(P<0.05,P<0.01)。结论:大黄虫丸加黄芪、水红花子后能改善TIMP-1、MMP-1的m RNA表达水平,抗大鼠肝纤维化作用更明显。
Objective: To investigate the effects of Dahuang Zhechong Pill mixtured with Huangqi and Shuihonghuazi on hepatic fibrosis of rats. Methods: Thirty-five SD rats were randomly divided into a normal group, model group, colchicine group, Dahuang Zhechong group, modified Dahuang Zhechong group. At the end of the experiment of 12 weeks, liver tissue samples were collected, pathological staining was used to evaluate the degree of rat hepatic fibrosis, RT-PCR technology was used to detect rat liver tissue matrix metalloproteinase-1(MMP-1), metalloproteinase inhibitor-1(TIMP-1) corresponding to the m RNA expression levels. Results: Compared with the normal group, m RNA of MMP-1 and m RNA of TIMP-1 had increased in model group(P〈0.01, P〈0.05); Compared with the model group, MMP-1 m RNA of Dahuang Zhechong group, modified Dahuang Zhechong group had increased and TIMP-1 m RNA decreased, in which modified Dahuang Zhechong group had significant difference(P〈0.01, P〈0.05). Conclusion: Dahuang Zhechong Pill mixtured with Huangqi and Shuihonghuazi can improve the expression of TIMP-1, MMP-1 m RNA with better effect on inhibition of rats with hepatic fibrosis.
