骨髓间充质干细胞治疗野百合碱诱导肺动脉高压大鼠肥大细胞的效果 被引量:1

Therapeutic effect of BM-MSCs on mast cells in MCT-induced PAH rats

摘要 目的:探讨骨髓间充质干细胞(BM-MSCs)移植治疗野百合碱(MCT)诱导的肺动脉高压(PAH)大鼠肥大细胞的效果。方法:雄性SD大鼠24只,随机均分为空白对照组、PAH组和BM-MSCs移植组。PAH组和BMMSCs移植组大鼠一次性腹腔注射MCT(60mg/kg)诱导PAH。采用SD大鼠的骨髓,经贴壁筛选法体外培养并纯化BM-MSCs。3周后,BM-MSCs移植组大鼠经右侧颈外静脉注射5×10~6个MSCs细胞悬液1ml,PAH组和空白对照组大鼠予等量L-DMEM液。移植后14d,观察比较三组大鼠的肺动脉收缩压(PASP)、右室肥厚指数(RVHI)、肺动脉的结构及肥大细胞数量的改变。结果:与空白对照组比较,PAH组和BM-MSCs组PASP[(23.75±0.83)mmHg比(46.73±1.28)mmHg比(35.54±1.20)mmHg]、RVHI[(0.2387±0.0048)比(0.4764±0.0155)比(0.3729±0.0087)]显著升高,肥大细胞计数[(0.63±0.26)个/10mm^2比(12.50±1.41)个/10mm^2比(7.13±0.64)个/10mm^2]显著增多,P均<0.01;但与PAH组比较,BM-MSCs组上述三项指标有显著降低,P均<0.01。肺组织病理切片显示PAH组肺血管重构明显,而BM-MSCs移植组肺血管重构明显减轻。结论:肥大细胞参与了肺动脉高压的形成,揭示了肺动脉高压发生的相关机制。 Objective:To explore therapeutic effect of bone marrow mesenchymal stem cells(BM-MSCs)transplantation on mast cells in rats with monocrotaline(MCT)induced pulmonary artery hypertension(PAH).Methods:A total of 24 male SD rats were randomly and equally divided into blank control group,PAH group and BM-MSCs transplantation group(BMMSCs group).PAH was induced in PAH group and BM-MSCs group through intraperitoneal injection of MCT(60mg/kg)once.Bone marrow of SD rats was cultured in vivo through adherence screening method and then purified to obtain BMMSCs.After three weeks,rats in BM-MSCs group received injection of 5×10~6 MSCs suspension via right external jugular vein while other two groups received L-DMEM solution of same volume.On 14 dafter transplantation,changes of pulmonary arterial systolic pressure(PASP),right ventricular hypertrophy index(RVHI),pulmonary artery structure and number of mast cells were observed and compared among three groups.Results:Compared with blank control group,there were significant rise in PASP[(23.75±0.83)mmHg vs.(46.73±1.28)mmHg vs.(35.54±1.20)mmHg],RVHI[(0.2387±0.0048)vs.(0.4764±0.0155)vs.(0.3729±0.0087)]and mast cell count[(0.63±0.26)/10mm^2 vs.(12.50±1.41)/10mm^2 vs.(7.13±0.64)/10mm^2]in PAH group and BM-MSCs group,P〈0.01all;compared with PAH group,there were significant reductions in above three indexes in BM-MSCs group,P〈0.01 all.Pulmonary pathophysiological section indicated that pulmonary vessels significantly reconstructed in PAH group,while it significantly relieved in BMMSCs group.Conclusion:Mast cells participate in formation of pulmonary artery hypertension,revealing related mechanism of pulmonary artery hypertension.
