血管生成相关因子内皮抑素和血小板反应素-1在多囊卵巢综合征大鼠卵巢组织中的表达及意义 被引量:6

摘要 目的探讨多囊卵巢综合征(PCOS)大鼠卵巢组织中内皮抑素(ES)和血小板反应素-1(TSP-1)的表达及意义。方法 2014年12月—2015年4月于哈尔滨医科大学附属第二医院动物实验中心进行实验。选取6周龄清洁雌性SD大鼠40只,随机分为对照组20只,模型组20只。模型组予来曲唑1 mg·kg^(-1)·d^(-1)连续灌胃21 d建立PCOS大鼠模型,对照组予等剂量1%羧甲基纤维索灌胃。造模成功后处死各组大鼠留取血清样本并摘取双侧卵巢组织取材,检测血清卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E_2)、孕激素(P)及睾酮(T)水平,观察大鼠卵巢形态学及组织学变化,并采用免疫组织化学法检测卵巢组织中ES和TSP-1的表达。结果模型组大鼠卵巢体积增大,可见包膜下囊性扩张的卵泡,镜下观察可见卵巢组织结构紊乱。与对照组比较,模型组大鼠体质量和卵巢质量均明显升高(t=3.918、2.389,P=0.000、0.024);模型组血清T、LH及E_2水平均增高,而FSH及P水平均降低(t=-2.309、2.870、5.330、-7.513、7.323,P<0.05或<0.01);模型组ES阳性(+++)分布显著高于对照组,而TSP-1阳性(+++)分布低于对照组(U=-4.762、-4.898,P=0.000、0.002)。结论来曲唑能够成功诱导PCOS大鼠模型,ES及TSP-1在PCOS的发生发展中发挥着重要作用。 Objective To discuss the expression and significance of endostatin(ES) and thrombospondin-1(TSP-1)of polycystic ovary syndrome(PCOS) rat model induced by letrozole.Methods The experiment was carried out in animal experiment center of second affiliated hospital of Harbin medical university from December 2014 to April 2015,forty female SD rats of six weeks age were randomly divided into control group(20 cases) and model group(20 cases).In model group,using Letrozole,with a dose of 1 mg·kg^-1·d^-1,continuing administered by Intragastric infusion for21 days to create PCOS model.After the success of the model,put the two groups of rats to death,and collect serum sample and remove bilateral ovarian tissue,the level of serum sex hormone such as follicle stimulating(FSH).luteinizing hormone(LH),estradiol(E2),progesterone(P) and T were detected and ovary morphology histological observation were observed under a microscope to evaluate model results,meanwhile,the expression of ES and TSP-1 in ovary were detected by the method of immunohistochemistry.Results In rats of model group,the ovarian volume increased,cystic expansion of the follicles could be seen under envelope,microscope showed ovarian tissue structure disorder.Compared with control group,the body mass and ovarian mass in rats of model group was in creased obviously(t=3.918,t=2.389,P=0.000,P=0.024).The levels of T.LH and E-2 were increased,while the levels of FSH and P was decreased(t=-2.309,t= 2.870,t= 5.330,t=- 7.513,t= 7.323,P〈0.05 or P〈0.01).The expression of ES in granular cells and theca cells was significantly higher in PCOS group than that in control group(Z=-4.762,P=0.000);The expression of TSP-1 in granular cells and theca cells was significantly lower in PCOS group than that in control group(Z=-4.898,P= 0.002).Conclusion Letrozole can induce rat model of PCOS successfully,the expression of ES and TSP-1 in the ovary is highly correlated with the pathogenesis of PCOS.
