摘要
目的对来自贵州省的两例布鲁菌病疑似病例进行布鲁菌分离与鉴定,为病例的确诊和疫情控制提供科学依据。方法采用虎红平板凝集试验及试管凝集试验检测布鲁菌病疑似病例布鲁菌抗体水平,采用血培养法对疑似病例进行细菌分离与培养.应用分子生物学技术结合传统方法对培养分离出的布鲁菌可疑菌株进行种型鉴定。结果自两名布鲁菌病疑似病例血液中分离出两株布鲁菌可疑菌株(命名为GZ13CB和GZ13ZSX)。布鲁菌表面蛋白31(BCSP31)属特异性PCR鉴定结果显示,两株菌出现布鲁菌特异的223bp预期扩增条带;AMOS.PCR种特异性PCR排除两株菌为羊种、绵羊附睾、猪种菌及牛种1、2、4型菌;进一步采用传统方法鉴定,两株菌均与牛种生物3型布鲁菌生物学特征相符。结论首次从布鲁菌疑似病例分离出牛种3型布鲁菌,此研究结果为贵州省牛种布鲁菌病疫情的控制提供了病原学依据。
Objective To etiologically diagnose and analyze two suspected patients of brucellosis and provide a etiologic basis for confirmation of the patients and control of brucellosis in Guizhou Province. Methods Antibody levels of Brucella in the suspected patients were determined by agar plate agglutination test and tube agglutination test. Blood culture method was used to isolate Brucella strains from the two suspected patients. Molecular and conventional techniques were used to identify suspicious bacterial strains isolated from the suspected patients. Results The results showed that two Brucella suspicious bacteria strains were isolated. Brucella genus specific surface protein 31 (BCSP31)-PCR showed a BruceUa abortus specific 223 bp-band in both of the two bacterial strains. Brucella species-specific AMOS-PCR excluded the two strains from BruceUa melitensis, BruceUa suis, BruceUa ovis, Brucella abortus biotype 1, 2 and 4. Conventional tests further identified the two strains as Brucella abortus biotype 3. Conclusion Two strains of Brucella abortus biotype 3 are firstly isolated from patients in Guizhou Province, which provides a etiological basis for diagnosis of the patients and prevention and control of brucellosis in Guizhou Province.
