摘要
目的探讨蒺藜皂苷对热损伤小鼠睾丸钠泵表达的影响。方法雄性小鼠30只,随机分为对照组、模型组和给药组,每组10只。建立睾丸热损伤模型,蒺藜皂苷灌胃(300 mg/kg,1 t/d,8 w)。一半小鼠取附睾精子进行精子参数检测;一半小鼠心脏灌流固定,进行免疫组织化学染色检测睾丸组织钠泵的表达。结果模型组与对照组比较,精子密度、活力、活率均显著下降,畸形率明显升高;生精细胞数目减少、排列紊乱;睾丸组织钠泵表达降低。给药组与模型组比较,精子密度、活力、活率升高,畸形率下降;生精细胞数目增多、排列紧密;睾丸组织钠泵表达升高。结论蒺藜皂苷能够通过调节钠泵表达对小鼠热损伤生殖功能的保护作用。
Objective To explore the effects of caltrop saponin on the expression of sodium pump in mice testis with thermal injury. Methods Thirty healthy male mice was randomly divided into control group, model group and treated group (n = 10). The thermal damaged model of the testis was established. The treated group was given sodium pump by intragastric administration(300 mg/kg) once a day for 8 weeks. Sperm parameters were detected in half of the mice by taking the epididymis sperm. The other half mice were fixed by perfusion through the heart. The expression of sodi- um pump in testis tissue was detected by immunohistochemical staining. Results Compared with the control group, the sperm density, the sperm motility and the percentage of live sperm were obviously decreased in the model group. And the rate of sperm deformation was obviously increased. The number of spermatogenic cells was reduced and the ar- rangement was in disorder. The expression of sodium pump in testis tissue was decreased. Compared with the model group, the sperm density, the sperm motility and the percentage of live sperm were obviously increased in the treated group. And the rate of sperm deformation was decreased. The number of spermatogenic cells was higher and the ar- rangement was in order. The expression of sodium pump in testis tissue was increased. Conclusion Caltrop saponin has protective effect on the damaged reproductive system in mice by regulating the expression of sodium pump.
