摘要 为建立快速、简便测定小麦茎基腐镰孢菌致病力的方法,将5mm的菌碟套接在催芽3d的小麦胚芽基部,接种后的小麦置于垫有湿滤纸的培养皿中,25℃下光暗(12h/12h)交替培养5d后测量麦苗的长度,以小麦苗长的降低率作为镰孢菌致病力的指标。结果发现,用该方法对12个菌株的致病力的测定结果与湿纸巾法有很强的相关性。经该方法测定,CF0915菌株的野生型和Tri5基因敲除突变体的致病力有显著差异。 A simple and rapid method for virulence assessment of Fusarium spp. causing crown rot of wheat was established. Three-day old wheat seedling was inoculated with the pathogen by inserting the whole germ through the center of a 5-ram mycelial plug; inoculated seedlings were put onto a Petri dish matted with water-saturated filter papers and cultivated at 25 ℃ with alternation of light and dark (12 h/12 h) for 5 d; the shoot length of each seedling was measured, and the reduction rate of wheat shoot length was taken as the indicator of pathogen virulence. Result showed that the virulence of twelve isolates assessed by this new method has strong relationship with that assessed by the method of wet-towel. Assessed by this new method, the virulence of wide type and Tri5-deletion mutant of isolate CF0915 showed significant difference.
