当归补血合剂对放射性肺炎小鼠血清转化生长因子和超氧化物歧化酶的影响 被引量:4

Effect of Danggui Buxue Decoction for transforming growth factor β1 and superoxide dismutase on radiation-induced pneumonia in mice

摘要 目的研究当归补血合剂对放射性肺炎模型小鼠血清转化生长因子(TGF-β1)、超氧化物歧化酶(SOD)的影响,以了解当归补血合剂对放射性肺炎的防治作用。方法将60只昆明小鼠随机分为4组:对照组、单纯给药组、单纯照射组、照射给药组;每组15只,建立急性放射性肺炎小鼠模型,以当归补血合剂灌胃给药,照射后21 d,检测TGF-β1以及SOD的含量变化;并制作小鼠肺组织切片行HE染色,观察小鼠肺组织病理改变。结果照射给药组血清TGF-β1浓度低于单纯照射组,照射给药组和单纯照射组TGF-β1的浓度均高于对照组和单纯给药组,差异有统计学意义(P<0.01);而照射给药组以及单纯照射组血清SOD的浓度低于对照组和单纯给药组,但照射给药组SOD浓度高于单纯照射组,差异均有统计学意义(P<0.01)。结论当归补血合剂可降低急性放射性肺炎小鼠的血清TGF-β1浓度,提高血清SOD的含量,在一定程度上减轻急性肺损伤,对放射性肺炎有一定的防护作用。 Objective To observe the influences of Danggui Buxue Decoction on transforming growth factor β1( TGF- β1) and superoxide dismutase( SOD) on radiation pneumonia mouse model,so as to reveal the impacts of Danggui Buxue Decoction on radiation- induced pneumonia. Methods 60 Kunming mice were randomly divided into four groups: blank control group,simple Danggui Buxue Decoction group,simple radiation group,radiation + Danggui Buxue Decoction group. Each group contained15 mice. We constructed the acute radiation- induced pneumonia mouse model by whole- thorax irradiation,which were given Danggui Buxue Decoction. After 21 days irradiation,all groups of mice were collected to detect the level of TGF- β1 and SOD.Fixed with formalin,the lung tissue was stained with HE,we examined the pathological changes of lung tissue. Results Compared with the radiation group,the serum concentration of TGF- β1 in radiation + Danggui Buxue Decoction group was lower than the simple radiation group,and the serum concentrations of TGF- β1 in both radiation + Danggui Buxue Decoction group and simple radiation group were higher than the control group and simple Danggui Buxue Decoction group,with the differences statistically significant( P 〈 0. 01). However,the concentrations of SOD in radiation + Danggui Buxue Decoction group and simple radiation group were lower than the control group and simple Danggui Buxue Decoction group,while the concentration of SOD was higher in the radiation + Danggui Buxue Decoction group than in the simple radiation group,with the differences statistically significant( P 〈 0. 01). Conclusion Danggui Buxue Decoction can decrease the serum concentration of TGF- β1 in acute radiation- induced pneumonia mice and increase the serum concentration of SOD,to some extent,it could alleviate the acute radiation- induced lung injury.
