摘要
在鸡1号染色体(GGA1)的Contig.060226.1上一个500 kb的区域选取30个SNP位点,研究江西省三个地方鸡种(宁都黄鸡、白耳黄鸡、安义瓦灰鸡)的单核苷酸多态性及连锁不平衡模式,估算其不同世代的群体有效规模大小。结果显示:在鸡驯养的较早期阶段(t=769),各品种的群体有效规模较大,宁都黄鸡、白耳黄鸡、安义瓦灰鸡分别为4 074、3 090、1 480;在鸡驯养的较晚期阶段,宁都黄鸡(t≤59)的群体有效规模为550,白耳黄鸡、安义瓦灰鸡(t≤45)分别为300、277。使用Sved模型对连锁不平衡系数r^2及传递的物理距离进行拟合,结果发现:宁都黄鸡、安义瓦灰鸡拟合状况良好(P=0.0272,R2=0.9196;P=0.0193,R^2=0.7276);白耳黄鸡虽未达到显著水平(P=0.3837,R^2=0.2541),但也表现出逐步递减的趋势,其r^2总变异方差的25.41%是由传递距离引起的。表明对宁都黄鸡、安义瓦灰鸡群体有效规模的估算相对准确,对白耳黄鸡群体有效规模的估算可能有偏。研究结果对于江西省三个地方鸡种特别是宁都黄鸡、安义瓦灰鸡的原位保存有重要的理论指导意义。
30 single nucleotide polymorphisms (SNPs)located in a 500 kb region within a Contig.060226.1 on GGA1 were selected to study the SNP diversity,linkage disequilibrium,and calculation of the effective population size (Ne) for three native chicken (Ningdu Huang chicken, Baier Huang chicken and Anyi Wahui chicken)in Jiangxi province.The results indicated that Ne was greater at the early stage of domestication (t=769),Ne of Ningdu Huang chicken, Baier Huang chicken and Anyi Wahui chicken were 4 074,3 090 and 1480,respectively. At the later stages of domestication, Ne of Ningdu Huang chicken was 550 (t≤59),and Ne of Baier Huang chicken and Anyi Wahui chicken were 300 and 277,respectively (t≤45). The Sved formula was fitted to the LD value (r2) and the physical distance. It was found that the regressions were significant for Ningdu Huang chicken (P=0.0272,R2=0.9196),and Anyi Wahui chicken (P=0.0193, R1=0.7276). Although the regression was not significant for Baier Huang chicken (P=O.3837,R2=0.2541),25.41% of the total variation of r2 could be explained by the transmitted physical distance. It was concluded that the estimation of Ne for Ningdu Huang chicken and Baler Huang chicken was accurate relatively,the estimation for Baier Huang chicken was more likely to be bias. This study had important theoretical significance in situ preservation for three native chickens, especially for Ningdu Huang chicken and Anyi Wahui chicken.
