摘要
目的应用基因表达谱芯片技术筛选过敏性紫癜患儿疾病发生及肾损伤相关基因,以期从分子水平探讨儿童过敏性紫癜发生发展及肾损伤发生机制。方法选取2010年1月至2010年12月在江西省儿童医院住院的无血缘关系的中国汉族过敏性紫癜患儿6例,根据有无肾脏损害分为单纯过敏性紫癜组3例(HSP组)和紫癜性肾炎组3例(HSPN组),另外选取同期在本院门诊体检且与HSP组和HSPN组性别、年龄相匹配的无血缘关系的中国汉族健康儿童3例作为对照组,分别检测各组患儿外周血基因表达谱情况,用相关生物信息学分析方法对不同实验组间基因表达谱进行差异分析。结果⑴单纯过敏性紫癜组与对照组间共有1064条差异表达基因,其中上调基因661条,下调基因403条,2倍以上差异基因225条,3倍以上6条;⑵紫癜性肾炎组与对照组间共有1326条差异表达基因,其中上调基因397条,下调基因929条,其中2倍以上差异基因619条,3倍以上103条;3、紫癜性肾炎组与单纯过敏性紫癜组间共有830条差异表达基因,其中上调基因445条,下调基因385条,2倍以上差异基因283条,3倍以上51条。4、2倍以上差异表达基因经GO分析,发现主要涉及炎症/免疫、细胞结构/功能、物质/能量代谢、信号转导/传递、转录/翻译等生物学过程。结论基因芯片能筛查出过敏性紫癜差异表达基因,为深入研究这些差异表达基因在过敏性紫癜中的作用提供了基础研究。
Objective To screen differentially expressed genes in children Henoch Schonlein purpura development and renal injury by c DNA microarray,and further analyze the pathogenesis of Children Henoch-Schonlein purpura development and renal injury from the molecular level.Methods Selected 6 patients with Henoch Schonlein purpura at Jiangxi Provincial Children's Hospital from January 2010 to December 2010,and they were divided into simple Henoch-Schonlein purpura group(HSP),Henoch-Schonlein purpura nephritis group(HSPN) according to the degree of renal damage.Also selected three healthy children as a control group.Detected the gene expression profiles of the peripheral blood of the three groups,and analyzed the differential of gene expression profiles between different experimental groups by relevant biological information,respectively.Results 1、The simple Henoch-Schonlein group compared with the control group:there are a total of 1064 differentially expressed genes,of which225 genes in more than two times and 6 genes more than 3 times;There are 661 genes up-regulated and 97 genes down-regulated in the differentially expressed genes.2、The Henoch-Schonlein purpura nephritis group compared with the control group:There are a total of 1326 differentially expressed genes,of which 619 genes in more than two times and 103 genes more than 3 times;There are 397 genes up-regulated and 929 genes down-regulated in the differentially expressed genes.3 、The Henoch-Schonlein purpura nephritis group compared with the simple Henoch-Schonlein purpura group:There are a total of 830 differentially expressed genes,of which 283 genes in more than two times and 51 genes more than 3 times;There are 445 genes upregulated and 385 genes downregulated in the differentially expressed genes.4、The more than two times differentially expressed genes were analyzed by the GO biology process analysis and were found that these genes were involved in five categories of biological processes inflammation/immunology,cell structure/functio
