摘要
目的 探讨肝X受体对急性肺损伤的保护机制,观察凋亡抑制因子Api6、Caspase-3、Bcl-2的表达情况.方法 将54只雄性SD大鼠随机分为正常对照组(CL组)、内毒素处理组(LP组)、T0901317预处理组(TP组).TP组预处理后,脂多糖造模,于1h、4h、8h时处死大鼠采取肺组织,免疫组化染色测定凋亡因子Caspase-3、Bcl-2,蛋白定量检测Api6表达水平,同时测定动脉血气,观察电子显微镜下肺组织病理学改变.结果 与CL组相比,LP组动脉血氧分压均显著降低;病理形态学示:与LP组相比,TP组损伤明显减轻.通过Western blot检测肺Api6蛋白表达获知:TP组明显比LP组表达有所增加.结论 Api6在ALI大鼠肺组织中表达明显减少.Api6高表达对肺组织起到一定的保护作用.肝X受体激动剂预处理可以减轻脂多糖所致肺组织的损伤,其机制可能与其抑制促凋亡因子、增加抗凋亡因子表达有关.
Objective To study the mechanism of liver X receptor protecting acute lung injury and to observe the expressions of apoptosis inhibiting factors—— Api6,Caspase 3,and Bcl-2.Methods 54 male SD rats were randomly divided into a normal control group (CL),an endotoxin treatment group (LP),and a T0901317 pretreatment group (TP).The TP group were made models with lipopolysaccharide after pretreatment.Hour 1,4,and 8,the rats were put to death and their lung tissue was collected.Caspase-3 and Bcl-2 were detected by immunohistochemical staining determination and Api6 by protein quantitative detection.The arterial blood gas was detected.The pathological changes of lung tissue were observed under electron microscope.Results The arterial blood oxygen partial pressure was significantly lower in the LP group than in the CL group.Pathological morphology showed that the injury was relieved more in the LP group than in the LP group.Western blot detection showed that the expression of Api6 was higher in the TP group than in the LP group.Conclusions The expression of Api6 significantly decreases in the lung tissue in ALI rats.The high expression of Api6 can protect lung tissue.Liver X receptor agonist pretreatment can reduce the lung tissue damage caused by lipopolysaccharide;its mechanism may be inhibiting apoptosis factors and increasing the expressions of antiapoptotic factors.
