摘要
目的:考察铁皮石斛对小鼠急性酒精性肝损伤的保护作用.方法:昆明小鼠随机分为正常组,模型组,铁皮石斛榨汁低、中、高剂量组[质量分数分别为0.5,1,2(g/kg)],当飞利肝宁组[DFLGN质量分数0.5(g/kg)].除正常组外,各组每天采用酒精灌胃造模,同时分别给予相应治疗药物.2周后测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,并观察肝组织形态学改变,应用Western Blot法检测肝组织凋亡通路p53和Bcl-2蛋白的表达情况.结果:与模型组比较,铁皮石斛干预后,血清ALT、AST水平显著下降(P<0.05);肝组织病理HE染色显示胞浆水样变性及气球样变有所改善;肝组织中抗凋亡蛋白Bcl-2显著上升(P<0.05).结论:铁皮石斛对急性酒精性肝损伤具有保护作用.
Aim：To determine the protection of Dendrobium Officinale （DO）against acute alcoholic liver injury in mice.Methods：KMmice were randomly divided into normal,model,positive control and three doses of Dendrobium Officinale juice groups.Except for those in normal group,mice were intragastrically infused with ethanol,and treated with water,DO [0.5,1 ,2（g/kg）],or DFLGN （0.5 g/kg）. At the end of the experiment,the protective effect was evaluated by determination of serum Aspartate transaminase （AST）and Alanine aminotransferase （ALT）,as well as observation of liver pathology by hematoxylin eosin （HE）staning,and detection on the expression levels of p53 and Bcl-2 protein in liver cells by Western Bloting.Results：Compared with model group,Dendrobium Officinale significantly reduced the levels of serum ALT and AST （P 〈0.05）.The pathology examination of liver showed that water cytoplasm degeneration and ballooning were improved.Additionally,the expression level of Bcl-2 protein was increased significantly （P 〈0.05）.Conclusion：Dendrobium Officinale can protect mice against alcoholic liver injury.
