红藻氨酸致癫痫大鼠不同时点神经生长因子变化研究

摘要 目的探讨红藻氨酸(KainicAcid,KA)致癫痫大鼠癫痫发作过程中海马组织神经生长因子(nervegrowthfactor,NGF)不同时间点表达量的变化。方法腹腔注射KA建立大鼠癫痫模型。采用TaqMan探针实时定量PCR(Real—timequantitativePCR)技术,检测注射KA后不同时点大鼠海马组织中NGF表达量的变化。结果与NGF表达量与生理盐水对照组(Ns)相比,6—12hNGF表达量明显低于Ns组(P〈0.05)、48—72hNGF表达量开始升高并高于NS组,在24、72h时其表达量显著高于NS组(P〈0.05)。结论NGF对致癫大鼠的海马具有修复与保护作用。 Objective To investigate the difference of expression of nerve growth factor(NGF) at different time by rats that induced epilepsy by kainic acid ( KA). Method The epilepsy model was used kainic acid intraperitoneal injectioned in rat. The nerve growth factor expression were detected by real-time fluorescent quantitative PCR assay with TaqMan probe in different time after intraperitoneal KA. ResuR Compared with normal sodium(NS) group, the expression of NGF were lower at 6-12 h , and were higher at 48 h and 72 h(P 〈0.05) ,but the expression of NGF were risied up at 48-72 h and higher by NS. Conclusion NGF have reparation and protection the hippocampus of rats that induced epilepsy by KA.
