摘要
目的掌握苏州市自然界蚊虫中携带乙型脑炎病毒情况。方法 2013年7月份在苏州市采集蚊虫样本,利用乙型脑炎病毒PreM/C基因特异性引物进行real-time-PCR(RT-PCR),阳性样本通过C6/36细胞分离乙型脑炎病毒,应用Clustal X和MEGA 6生物软件完成阳性样本核苷酸同源性分析和进化树构建。结果共采集蚊虫1 055只,在三带喙库蚊中检出GⅠ型乙型脑炎病毒1株。结论首次证实苏州市蚊虫携带基因Ⅰ型乙型脑炎病毒,应在以后的监测和日常生活中加强防范。
Objective To find out the infection rate of Japanese encephalitis virus from mosquitoes in Suzhou city. Methods Mosquito samples were collected in Suzhou city in the summer of 2013, and then RT-PCR was used to identify the positive isolates using PreM/C gene specific primers for Japanese encephalitis vi- rus. The positive samples were inter-cells cross cultured for viral isolation. The molecular biological analysis was carried out by Clustal X software and MEGA 6. Results A total of 1 055 mosquitoes were collected at Suzhou city and the isolated Japanese encephalitis viruses from these mosquitoes were identified to be genotype 1. Conclusions This study confirmed genotype 1 of Japanese encephalitis virus from mosquitoes for the first time in Suzhou city. Monitoring and prevention are needed in future.
