摘要
目的研究Rho蛋白鸟苷酸交换因子11(ARHGEF11)基因R1467H多态性与多囊卵巢综合征(PCOS)及其胰岛素抵抗(IR)的关系。方法应用聚合酶链反应-限制性片段长度多态性技术对136例PCOS患者和117名健康对照者ARHGEF11基因R1467H位点进行基因分型;同时进行人体测量学和代谢指标的检测。结果在PCOS组和对照组中,ARHGEF11基因R1467H位点的基因型和等位基因频率分布差异无统计学意义(P>0.05);PCOS组GA/AA基因型的空腹胰岛素和HOMA2-IR水平明显高于GG基因型(P<0.05),对照组GA/AA基因型的空腹血糖显著高于GG基因型(P<0.05),这种统计学差异经年龄、体质量指数校正后依然存在(P<0.05)。结论 ARHGEF11基因R1467H多态性与PCOS的发病无关,但可能与PCOS患者IR抵抗有关。
Objective To study the association between Rho guanine nucleotide exchange factor 11gene(ARHGEF11)R1467H polymorphism and polycystic ovary syndrome(PCOS).Methods The single nucleotide polymorphism of ARHGEF11R1467 H was detected in 136 subjects with PCOS and 117 normal subjects by PCRrestriction fragment length polymorphism(PCR-RFLP).At the same time,the anthropometry and metabolism indices were measured.Results The distribution of genotype and allele frequency were not significantly different between the normal subjects and subjects in PCOS group(P〉0.05).In PCOS group,the fasting plasma insulin and HOMA2-IR of GA/AA genotype were higher than GG genotype(P〈0.05),the fasting plasma glucose of GA/AA genotype in control group was higher than GG genotype(P〈0.05).These differences were also statistically significant even after the adjustment for age and body mass index(P〈 0.05).Conclusion ARHGEF11R1467 Hpolymorphism isn't associated with genetic susceptibility of PCOS,but it may be related to its insulin resistance.
