摘要
目的 观察白藜芦醇抑制白细胞介素(IL)-6诱导的体外SGC7901细胞侵袭的作用,探讨其作用机制.方法 采用噻唑蓝(MTT)法确定白藜芦醇对SGC7901细胞无明显毒性的浓度范围,并以该浓度范围内的白藜芦醇作用IL-6诱导的SGC7901细胞,Transwell小室法检测其抑制细胞侵袭作用,明胶酶谱法分析细胞的基质金属蛋白酶(MMP)-2活性变化,Western blot检测IL-6信号通路相关蛋白表达.结果 30 μmol/L内的白藜芦醇无明显的细胞毒性,20 μmol/L处理的受IL-6诱导的SGC7901细胞侵袭能力明显降低,约为20%,MMP-2分泌减少约为50%,细胞内丝裂原活化蛋白激酶(MAPK)通路的相关分子磷酸化细胞外信号调节激酶1/2 (p-ERK1/2)含量下降.结论 20 μmo]/L白藜芦醇可有效抑制SGC7901细胞侵袭,其机制可能是通过抑制ERK通路相关蛋白激活,最终降低MMP-2的分泌.
Objective To elucidate the inhibitory effect of resveratrol on interleukin (IL)-6-induced invasive ability of SGC7901 cells and the molecular mechanism.Methods The cell viability of cells treated with resveratrol was evaluated by methyl thiazolyhetrazolium (MTT) assay.The SGC7901 cells were treated with IL-6 and non-cytotoxic concentration of resveratrol.The invasion of SGC7901 cells was detected by Boyden chamber assay.Matrix metalloproteinase (MMP)-2 activity was determined by gelatine zymography assay.The changes of the related proteins were detected by Western blotting.Results Resveratrol was not toxic to SGC7901 cells at the concentration under 30 μmol/L.The invasion ability of IL-6-induced SGC7901 cells was decreased about 20% after treatment with 20 μmol/L resveratrol for 24 h,accompanied by the inhibition of MMP-2 secretion for 50%,as well as the levels of phosphorylated extracellular regulated protein kinases 1/2 (p-ERK1/2).Conclusion Resveratrol of 20 μmol/L inhibits invasion of SGC7901 cells possibly through the suppression of the activation of ERK signaling pathway,subsequently exerting inhibitory effect on MMP-2.
