摘要
从城市污水处理厂市政污泥中回收蛋白质是一种污泥资源化的新途径,等电点沉淀法是从市政污泥水解液中回收蛋白质的有效方法。结果表明,当pH为3.5、沉淀温度为5℃、离心转速为3 000 r·min^(-1)和蛋白质浓度为6 000 mg·L^(-1)时,市政污泥水解液中的蛋白质回收率为87.46%。采用高效液相色谱仪分析所得蛋白质产品中的氨基酸含量,发现7种必需氨基酸(赖氨酸、苯丙氨酸、蛋氨酸、苏氨酸、异亮氨酸、亮氨酸和缬氨酸)占沉淀物干重的12.37%。采用电感耦合等离子体发射光谱仪(inductively coupled plasma optical emission spectrometer,ICP-OES)分析所得蛋白质的重金属含量,与原市政污泥相比,所得蛋白质中As、Pb、Cr等重金属的削减率均在90%以上,等电点沉淀法有利于市政污泥中重金属的去除,所得蛋白质产品存在重金属风险较低。
Protein recovery from the waste sludge of municipal wastewater treatment plants is one of the new approaches to sludge resource recovery. The isoelectric point precipitation method is an efficient way to recover proteins from municipal sludge. The results of this study showed that the protein recovery rate was as high as87. 46% at a pH of 3. 5,precipitation temperature of 5 ℃,centrifugal speed of 3 000 r·min-1,and protein concentration of 6 000 mg·L-1. The amino acid content of the obtained sludge protein was analyzed by high performance liquid chromatography. It was found that seven essential amino acids( lysine,phenylalanine,methionine,threonine,isoleucine,leucine,and valine) accounted for 12. 37% of the protein recovery rate. The heavy metal content of the obtained sludge protein was analyzed by inductively coupled plasma optical emission spectroscopy( ICP-OES),demonstrating that the removal of As,Pb,and Cr was more than 90% compared with that of the original municipal sludge. The isoelectric point precipitation method proved to be beneficial for the removal of heavy metals in municipal sludge,reducing the risk of heavy metal pollution in the protein product.
