摘要
目的:探讨慢性丙型肝炎病毒(HCV)感染者白细胞介素28B(IL28B)基因多态性的分布特点及其与HCV-RNA载量、肝功能之间的关系。方法:对94例慢性HCV感染者进行IL28B(rs8099917、rs12979860、rs12980275)基因多态性、HCV-RNA载量及肝功能检测,比较IL28B基因型与HCV-RNA载量及肝功能的关系。结果:94例患者中,rs8099917位点TT基因型82例(87.2%)、TG基因型12例(12.8%)、GG基因型0例;rs12979860位点CC基因型78例(83.0%)、CT基因型16例(17.0%)、TT基因型0例;rs12980275位点AA基因型75例(79.8%)、AG基因型19例(20.2%)、GG基因型0例。IL28B基因多态性的分布与HCV-RNA载量无显著性相关(P>0.05);仅rs12979860位点与血清丙氨酸氨基转移酶(ALT)水平有关,CC基因型者血清ALT水平高于CT基因型者,差异有统计学意义[(66.9±51.2)U/L vs(44.7±21.6)U/L,P=0.008]。结论:慢性HCV感染者IL28B基因rs8099917、rs12979860、rs12980275位点分别以TT、CC、AA为主;以上3个位点的变异与慢性HCV感染者的HCV-RNA水平均无关;而rs12979860CC基因型者具有较高的ALT水平。
Objective To investigate the distribution of (interleukin 28B) IL28B gene single nucleotide poly-morphism (SNP) genotypes in patients infected with chronic hepatitis C virus (HCV) and its clinical sig-nificance. Methods: A total of 94 patients infected with HCV were enrolled in this study. IL28B geincluded rs8099917, rs12979860 and rsl 2980275 were identified and were compared in HCV patients to as-sess the influence of SNP on HCV-RNA viral load and liver function. Results: The prevalence of rs8099917 TT, TGand GG were 82 (87.2%), 12 (12.8%) and 0 (0%), respectively. rs12979860 CC, CT and TT were 78 ( 83.0%), 16 (17.0%) and 0 (0%), respectively. rs12980 275 AA, AG and GG were 75 (79.8%), 19 (20.2%) and 0 (0%) , respectively. There was no relationship between IL28B SNPs and HCV-RNA viral load ( P 〈0. 05). The serum level of alanine aminotransferase (ALT) in the patients who carried rsl 2979860 CC was signigicantly higher than those who carried rs1 2979860 CT [CC CT, (66. 9 ± 51. 2) U/L (44. 7 ± 21. 6) U /L ,P = 0. 008]. Conclusion: Most of the patients infected with HCV carry IL28B genotypes rs8099917 TT, rs12979860 CC and rs1 2980275 AA. And the variation of the above three genotypes was not correlated with HCV-RNA level. Patients who carried s1 2979860 CC had a higher serum level of ALT than those who carried rs12979860 CT.
