摘要
目的:研究淫羊藿和女贞子配伍对激素局部干预哮喘大鼠肺热休克蛋白90(HSP90)、糖皮质激素受体(GR)的影响作用。方法:SPF级雄性SD大鼠,随机分为正常对照组(N组)、哮喘模型组(A组)、布地奈德组(GC组),淫羊藿女贞子组(YN组),淫羊藿女贞子合布地奈德组(YN+GC组)。卵蛋白致敏及激发建立大鼠哮喘模型,YN+GC组以布地奈德雾化4周治疗,同时配合灌胃淫羊藿女贞子煎液;GC组、YN组分别单用相应药物治疗。免疫细胞化学检测肺泡灌洗液(BALF)中HSP90蛋白表达,免疫组织化学法检测肺组织HSP90蛋白表达,实时定量PCR法检测肺组织HSP90及GR的m RNA表达。结果:与N组比较,A组大鼠肺组织HSP90蛋白、GRαm RNA及GRα/HSP90m RNA比值显著降低(P<0.01,P<0.05);与A组比较,YN组显著上调肺GRαm RNA表达(P<0.01),YN+GC组上调肺HSP90蛋白及m RNA与GRαm RNA表达(P<0.05,P<0.01);与GC组比较,YN+GC组可显著上调BALF及肺组织中HSP90蛋白表达,肺HSP90及GRαm RNA表达及GRα/HSP90m RNA比值(P<0.05,P<0.01)。结论:改善GR-HSP90作用可能是平补阴阳药对淫羊藿和女贞子协同激素局部给药发挥作用的机制之一。
Objective： To research the combination of Herba Epimedii and Fructus Ligustri Lucidi on HSP90-GR in the asthmatic rats with Budesonide intervention. Methods： Male SD rats were randomly divided into the control group（N group）, asthma model group（A group）, Budesonide group（GC group）, combination of Herba Epimedii and Fructus Ligustri Lucidi group（YN group）, and combination of Herba Epimedii and Fructus Ligustri Lucidi group accompanied by the atomization inhalation of Budesonide group（YN＋GC group）. The rat model of asthma was induced by ovalbumin（OVA）. The Budesonide was administration by inhalation for 4 weeks, while Herba Epimedii and Fructus Ligustri Lucidi Decoction was given by gavage. The protein expression of HSP90 in bronchoalveolar lavage fluid（BALF） and lung tissue was detected respectively by immunocytochemistry and immunohistochemical method. And the expression of HSP90 and GR m RNA in lung tissue were detected by RT-PCR. Results： Compared with the N group, the protein expression of HSP90, the m RNA expression of GRαand the ratio of GRα m RNA to HSP90 m RNA in the lung tissue were decreased significantly by A group（P〈0.01, P〈0.05）. Compared with A group, the m RNA expression of GRα of YN group was increased significantly（P〈0.01）, and m RNA expression of HSP90 and GRα and the protein expression of HSP90 of YN＋GC group were increased significantly（P〈0.05, P〈0.01）. Compared with GC group, the protein expression of HSP90 in BALF and lung tissue, the m RNA expressions of HSP90 and GRα and the ratio of GRα m RNA to HSP90 m RNA in the lung tissue of the YN＋GC group were increased significantly（P〈0.05, P〈0.01）. Conclusion： The synergistic action of Herba Epimedii and Fructus Ligustri Lucidi combined with Budesonidein the treatment of asthma may be related to the improvement of GR-HSP90.
