摘要
通过自制过滤装置,研究了酶-重量法的最佳测定条件,测定了燕麦中不可溶性膳食纤维及可溶性膳食纤维的含量。实验结果表明该法最优化条件为:除酯用石油醚量为30ml,除酯超声时间为15min-25 min,α-淀粉酶酶解温度为70℃,酶解时间为30min,蛋白酶酶解p H7.5±0.2,葡萄糖苷酶p H4.0-5.0。结论:α-淀粉酶酶解时间、α-淀粉酶酶解温度、蛋白酶及葡萄糖苷酶酶解p H对测定结果影响较大,加酶量大于5mg时对测定结果基本无影响,石油醚用量对IDF及SDF含量测定有不同程度影响,但对SDF影响显著,超声时间大于15min对SDF含量测定影响较大,对IDF含量测定影响不大。结果表明该测量条件用于燕麦膳食纤维含量的测定结果较为满意。该法将为准确测定膳食纤维含量提供一定的实验依据。
Through self - filtering device to study the enzymatic - gravimetric method for determination of the optimum conditions for the determination of the content of the non - oat soluble dietary fiber and soluble dietary fiber. Conclusion: α - amylase hydrolysis time, α - amylase hydrolysis temperature, protease and glucosi- dase enzyme pH greater impact on the measurement results, when the enzyme is greater than 5mg no influence on the measurement results, determination of the amount of petroleum ether have different influences on the IDF and SDF content, but a significant effect on the SDF, ultrasonic longer than 15min to SDF determination greater impact on the determination of IDF little effect. The results show that the method used to determine the results of oat dietary fiber content is satisfactory. This method will provide an accurate de- termination of the dietary fiber content of some experimental basis.
