摘要
以金针菇(Flammulina velutipes)基因组序列为基础,对其SSR位点进行分析,设计16对引物对其中16个SSR位点进行标记设计,并对6个商业化的金针菇品种进行筛选。统计结果表明,金针菇基因组有1279个位点,SSR位点密度为35.53/Mb,并以三核苷酸基序为主,重复数≥10的SSR(即高频)基序数目总计246个,占所有SSR位点的19.23%。设计的16对SSR标记均能扩增条带,多态率仅为18.75%。
We have investigated SSR loci in the whole genome sequence of Flammulina velutipes and have developed 16 SSR markers for polymorphism analysis.A total of 1279 SSR loci were detected with an overall density of 35.53SSR/Mb Trinucleotide repeats were the most frequent motif in the genome(~60%)followed by mononucleotide(18.1%)and dinucleotide(16.1%)repeats.Altogether,246 high frequency(≥10)SSRs were identified,accounting for 19.23% of the total loci.All 16 SSR markers were successfully amplified in six cultivated strains of F.velutipes,and the polymorphism rate was 18.75%.
