尼克酰胺腺嘌呤氧化酶4在神经胶质瘤的表达

摘要 目的探讨尼克酰胺腺嘌呤氧化酶4(NOX4)在胶质瘤的可能作用。方法分析85例行手术切除并经病理证实的胶质瘤标本,WHOⅠ级、Ⅱ级、Ⅲ级和Ⅳ级分别12例、29例、10例和34例。应用免疫组化方法评估NOX4在各级别神经胶质瘤的表达,分析NOX4在胶质瘤组织的表达部位及表达量变化。结果 NOX4在胶质瘤的表达量较瘤旁组织明显增高。NOX4在低级别胶质瘤中主要表达于细胞质,在高级别胶质瘤的细胞质及细胞核均有表达。NOX4在肿瘤新生血管呈强阳性表达;NOX4在胶质瘤的表达量与胶质瘤恶性程度有关,恶性程度越高,NOX4表达量越高。结论 NOX4在胶质瘤过表达可能在胶质瘤发生和发展过程中起到重要作用。 Objective To investigate the possible role of nicotinamide adenine oxidase 4 (NOX4) in glioma. Methods Eighty-five glioma specimens from surgical resection and confirmed by pathology were analyzed. According to the World Health Organization (WHO) classification, Grade Ⅰ, Ⅰ/,Ⅲ and Ⅳ were were seen in 12, 29, 10 and 34 patients respectively. The expression of NOX4 in different grtade gliomas was assessed by immunohistochemical methods, the expression site and expression level of NOX4 in glioma tissue were analyzed. Results The expression of NOX4 was significantly higher in glioma than in the tumor-adjacent tissues. NOX4 was expressed mainly in the cytoplasm of low-grade glioma, and in the cytoplasm and nucleus of high-grade glioma. The expression of NOX4 was very strong in tumoral newly developed vessels. The expression level was positively related to the malignancy grade. Conclusion NOX4 overexpression in glioma cells may play an impotent role in the tumorigenesis and progression of glioma.
