全反式维甲酸联合干扰素对滤泡状甲状腺癌细胞摄碘的促进作用 被引量:2

摘要 目的通过药物诱导,提高甲状腺癌细胞对131I的摄取能力,是改善失分化甲状腺癌的有效方法之一。文中探讨全反式维甲酸(all trans retinoic acid,ATRA)联合干扰素(interferon,IFN)对滤泡状甲状腺癌细胞摄碘的促进作用。方法为比较单独用药与联合用药对甲状腺癌细胞摄碘率的影响,实验分为空白对照组、2μmol/L ATRA组(ATRA单药组)、1500IU/m L IFN-α2a组(IFN-α2a单药组)以及2μmol/L ATRA+1500 IU/m L IFN-α2a联合用药(联合用药组)诱导FTC-133细胞72 h后,Na125I摄取实验检测药物诱导后细胞摄碘变化、Na125I流出实验测定碘在细胞内的滞留率。结果与联合用药组FTC-133细胞摄碘(2 423.1±237.5)比较,空白对照组(1 279.5±102.8)、ATRA单药组(1 438.2±149.6)及单独用药IFN-α2a组(1 355.3±198.4)显著降低(P<0.05)。空白对照组、ATRA单药组、IFN-α2a单药组、联合用药组诱导后,Na125I在FTC-133细胞的滞留率组内各时间点差异有统计学意义(P<0.01),同一时间点各组间差异无统计学意义(P>0.05)。结论ATRA联合IFN-α2a有效增强FTC-133细胞摄碘能力,但不能提高碘在细胞内的滞留率。 Objective Some drugs can effectively improve the uptake of ^131I in dedifferentiated thyroid cancer. This study was to investigate the effect of all-trans retinoic acid (ATRA) combined with interferon-α2a (IFN-α2a) on the iodine uptake and retention rate of FTC-133 follicular thyroid carcinoma cells. Methods FTC-133 cells were cultured in the presence of 2μmol-/L AT- RA and/or 1500 IU/mL IFN-α2a for 72 hours. The the cells were collected for measurement of the uptake and retention rate of Na^125 I. Results The Na125I uptake of the FTC-133 cells was significantly increased after 72-hour incubation with 2 μmol/L ATRA and 1500 IU/mL IFN-α2a (2 423.1 ±237.5 ) as compared with those of the control ( 1 279.5 ± 102.8 ), ATRA ( 1 438.2 ± 149.6 ), and IFN-α2a groups ( 1355.3 ± 198.4) ( P 〈 0.05 ). Statistically significant differences were observed in the retention rate of Na^125I in the FTC-133 cells at different time points in the each of the blank control, 2 μmol/L ATRA, 1500 IU/mL IFN-α2a, and combination groups ( P 〈 0.01 ), but not in that of Na^125I among these groups ( P 〈0. 05 ). Conclusion ATRA in combination with IFN-α2a can promote the Na125I uptake but cannot improve the Na^125I retention rate in FTC-133 cells.
