摘要
为研究低温胁迫对木薯叶片抗氧化酶表达水平的影响,运用蛋白质印迹技术,选用中国主栽木薯品种华南205(SC205)组培苗为材料,研究了5℃低温胁迫7 d期间叶片的抗坏血酸过氧化物酶(APX)、谷胱甘肽还原酶(GR)、超氧化物歧化酶(SOD)和过氧化物酶(POD)4种酶表达水平的动态变化。结果表明:胁迫前期,随着胁迫时间的延长,4种酶的表达水平均呈上升趋势,SOD及GR在胁迫3 d时表达量最高,APX在胁迫5 d时表达量达到最高,POD在胁迫4 d的表达水平最高。达到最高值后,APX及SOD表达水平呈下降后又上升趋势,而GR及POD的表达水平则一直下降。该研究结果和方法可为进一步认识木薯耐低温的生理机制提供参考。
In order to study the effect of low temperature stress on expression levels of antioxidant enzymes in cassava leaves, cassava cultivar South China 205 ( SC205 ) were used as the materials, expression level of ascorbate peroxidase (APX) , glutathione reductase (GR) , superoxide dismutase (SOD) and peroxidase (POD) in leaves during 7 d after exposed to 5 ℃ were meas- ured by Western-blotting technique. The results showed that: in the early phase, the expression levels of 4 kinds of enzymes were increased with stress time, SOD and GR expression was the highest at stressed for 3 d, while APX was at 5 d and POD was 4 d. After reaching the highest, APX and SOD expression levels were fall after rising, while GR and POD was declined. The results and method can provide a reference for further understanding of the physiological mechanisms on low temperature tolerant in cassava.
