摘要
目的:探讨IL-1基因多态性与寻常型银屑病表型及其NFκB表达的相关性。方法:选取2012年1月至2015年9月我院收治的寻常型银屑病患者112例为研究对象,分析其皮损中IL-1β和IL-1RA的基因多态性,统计银屑病表型,同时采用免疫组化法检测其皮损中NFκB表达水平,比较不同IL-1基因多态性患者的银屑病表型及其皮损中NFκB表达水平,并采用Logistic多元回归分析IL-1基因多态性与寻常型银屑病表型及其NFκB表达的相关性。结果:112例寻常型银屑病中,IL-1β的CC、CT和TT基因型比例分别为16.96%、68.75%和14.29%,IL-1RA的AA、AB和BB基因型比例分别为61.61%、31.25%和7.14%。IL-1RA的AA基因型中NFκB阳性表达率高于AB型,而AB基因型又高于BB基因型(P值均<0.05)。IL-1β各基因型中NFκB阳性率无明显差异,斑块型和点滴型的患者比例亦无明显差异(P值均>0.05)。IL-1RA的AA基因型中斑块型患者比例高于AB型,而AB型又高于BB基因型患者(P值均<0.05)。Logistic多元回归分析结果显示,IL-1RA基因多态性与寻常型银屑病表型及NFκB表达均密切相关(P值均<0.05)。结论:IL-1RA基因多态性与寻常型银屑病表型及NFκB表达水平均相关,NFκB和IL-1RA基因可能共同影响寻常型银屑病的发生发展。
Objective :To explore correlation between IL-1 gene polymorphism with the psoria- sis phenotype and NlrKB positive expression rate. Methods: 112 cases of psoriasis patients in our hospital from January 2012 to September 2015 were selected as the research object. The IL-1β and IL-1RA gene polymorphisms were detected and immunohistochemical method was applied to detect the expression level of NFkB. Psoriasis phenotype and NFkB expression rate in patients with dif- ferent IL-1 gene polymorphism were compared and Logistic multivariate regression analysis method was applied to analyze correlation between IL-1 gene polymorphism with the psoriasis phenotype and NFkB positive expression rate. Results: IL-1β CC, CT and TT genotype ratio of the patients were 16.96%, 68.75% and 14.29% respectively, IL-1RA AA, AB, and BB genotype ratio were 61.61%, 31.25% and 7.14% respectively. NFkB positive expression rate of patients with IL-1 RA AA genotype was higher than that of patients with IL-1RA AB and BB genotype, and NFKB positive expression rate of patients with IL-1 AB genotype was higher than that of patients with IL- l RA BB genotype ( All of the P value were lower than 0.05 ). Proportion of patients with plaques and drop psoriasis phenotype of patients with IL-1 RA AA genotype was higher than that of patients with IL-1RA AB and BB genotype, Proportion of patients with plaques and drop psoriasis pheno- type of patients with AB genotype was higher than that of patients with IL-1RA BB genotype ( All of the P value were lower than 0.05 ). Logistic multivariate regression analysis results showed that the IL-1RA gene polymorphism and psoriasis phenotype, NFkB expression were closely related. Conclusion: IL-1 RA gene polymorphism and psoriasis phenotype, NFkB were closely correlated, therefore NFkB and IL-1 gene may synergistically promote psoriasis occurrence and progress.
