摘要
目的 探讨米非司酮诱导的早产大鼠模型外周血中IL-17的表达水平及其意义。方法SPF级10-12周龄Sprague-Dawley大鼠适应性喂养1周后雌雄合笼,阴道涂片发现精子者标记为妊娠第1日。将8只孕鼠随机分为对照组与早产组,每组4只。早产组大鼠在妊娠第18日皮下注射米非司酮10 mg/kg,建立米非司酮诱导的早产大鼠模型,对照组大鼠在相同时间注射芝麻油,分别于注射后6 h及24 h采集2组大鼠外周血,检测血清中IL-17、粒细胞-巨噬细胞集落刺激因子(GM-CSF)的水平。比较早产组和对照组间IL-17、GM-CSF水平的差异。结果 早产组大鼠在妊娠第18日皮下注射米非司酮后的发动分娩时间间隔明显短于皮下注射芝麻油的对照组(P〈0.01)。早产组大鼠在注射米非司酮后6 h时血清IL-17水平与对照组比较差异无统计学意义(P〉0.05),24 h时血清IL-17水平高于对照组(P〈0.01)。2组大鼠在注射米非司酮后6、24 h时血清GM-CSF水平比较差异均无统计学意义(P均〉0.05)。结论 米非司酮诱导的早产大鼠外周血中IL-17水平升高,提示炎症因子IL-17可能参与了激素诱导的早产发生过程。
Objective To investigate the expression level and significance of interleukin ( IL) -17 in the peripheral blood of mifepristone-induced preterm labor rat models. Methods Specific pathogen free (SPF) -grade Sprague-Dawley rats, aged 10-12 weeks, were acclimated for 1 week and then the male and female rats were housed and mated. A positive vaginal smear for sperm after mating was designated as gestation day 1 of pregnancy. Eight pregnant rats were randomly divided into control and preterm labor groups ( n = 4 for each group). In the preterm labor group, mifepristone-induced preterm labor rat models were established by subcutaneous injection of mifepristone at a dose of 10 mg/kg on gestation day 18 of pregnancy. In the control group, rats were injected with vehicle (sesame) on gestation day 18 of pregnancy. At 6 h and 24 h after injec-tion ,peripheral blood samples were collected for detection of the serum levels of IL-17 and granulocyte macro-phage-colony stimulating factor (GM-CSF). The serum levels of IL-17 and GM-CSF were statistically compared between two groups. Results After subcutaneous injection of mifepristone on gestation day 18 of pregnancy, the time interval of labor onset in the preterm labor group was significantly shorter compared with that in the control group (P 〈0. 01). At 6 h after administration of mifepristone, the IL-17 levels did not significantly differ between two groups (P 〉0. 05). At 24 h after administration of mifepristone, the IL-17 level in the preterm labor group was significantly up-regulated than that in the control group ( P 〈 0. 01 ). At 6- and 24-h after administration of mifepristone, the GM-CSF levels did not significantly differ between two groups (both P 〉0. 05).Conclusion The IL-17 level in the peripheral blood of mifepristone-induced preterm labor rats was significantly up-regulated, suggesting that IL-17, as an inflammatory cytokine, probably participates in the incidence of hormone-induced preterm
