摘要
目的:探讨三种冰片对大鼠肠道细胞色素P450和糖蛋白P-gp的影响。方法:分别口服灌胃三种冰片400mg/kg连续5天,通过制备肠微粒体、蛋白含量测定以及紫外分光光度法研究上述龙脑成分对肠段P450含量的影响。分别口服灌胃三种冰片200mg、400mg、800mg,在不同时间肠段取材,以组化染色的方法比较空肠和回肠中细胞色素CYP3A4和P糖蛋白的差异。结果:左旋龙脑和右旋龙脑能显著增加空肠回肠段P450的含量,左旋龙脑400mg和800mg能显著增加空肠和回肠中的CYP3A4,右旋龙脑200mg、400mg、800mg均能显著增加空肠和回肠中的CYP3A4,右旋龙脑400mg降低回肠中的P-gp,右旋龙脑800mg能降低空肠中的P-gp。合成冰片200mg、400mg、800mg均显著降低空肠和回肠中的CYP3A4和P-gp。结论:三种冰片对CYP3A4和P-gp的作用具有显著差异。
Objective: To investigate the effects of three kinds of borneol on rat intestinal cytochrome P450 and glycoprotein P-gp. Methods: Three kinds of bomeol were given orally respectively to rats in dose of 400mg for 5 days , then study on the effect of box'ned on P450 in intestinal with UV spectrophotometry . Three kinds of borneol were given orally to rats in dose of 200mg,400mg,800mg respectively and compared CYP3A4 and P-gp in jejunum and ileum with immunohistochemical staining. Results: L-borneol and D-borneol can significantly increase con- tent of P450 in jejunum and ileum. L-borneo1400mg and 800rag can significantly increased the jejunum and ileum of CYP3A4,D-borneol 200 mg 400mg and 800mg can significantly increased the jejunum and ileum of CYP3A4, D-borneol 400mg decreased in the ileum of P-gp, D-bor- neol 800mg can reduce in jejunum of P-gp. Synthetic borneo1200 mg,400 mg and 800 mg were significantly decreased in the jejunum and il- eum of CYP3A4 and P-gp. Conclusion: The effect of three kinds of borneol on CYP3A4 and P-gp has significant differences.
