摘要
目的探讨中药复方缬芎滴丸对大鼠脑缺血半暗带组织神经巢蛋白(Nestin)、双肾上腺皮质激素(DCX)、神经核蛋白(NeuN)表达的影响。方法 SD大鼠随机分为假手术组、模型组、缬芎滴丸(50mg/kg)给药组,线栓法制作大鼠大脑中动脉栓塞模型(MCAO),缺血30min再灌注,假手术组大鼠只结扎颈总动脉,不插栓。造模成功后对缬芎滴丸组大鼠灌胃给药,假手术组和模型组给予等量的蒸馏水。给药7d后采用免疫荧光染色技术观察缬芎滴丸对大鼠脑缺血半暗带组织Nestin、DCX、NeuN表达的影响。结果大鼠脑缺血半暗带组织Nestin表达量在假手术组为(18.67±0.58),模型组为(49.00±5.29),缬芎滴丸组为(105.67±3.51),给药组和模型组比较差异有统计学意义(P<0.01);DCX表达量在假手术组为(21.67±1.53),模型组为(61.00±4.36),缬芎滴丸组为(112.00±12.53),给药组和模型组比较差异有统计学意义(P<0.01);NeuN表达量在假手术组为(28.67±6.51),模型组为(46.00±4.36),缬芎滴丸组为(323.00±10.82),给药组和模型组比较差异有统计学意义(P<0.01)。结论缬芎滴丸可能通过促进大鼠脑缺血半暗带组织Nestin、DCX、NeuN表达,上调内源性神经发生,从而参与脑组织损伤修复过程。
Objective To investigate the effects of valerian ligusticum pill on the expression of Nestin,DCX,NeuN in the cerebral ischemic penumbra of rats.Methods SD rats were randomly divided into sham-operated group,model group,and valerian ligusticum pill(50mg/kg)group.The animal model was established by occlusion of the middle cerebral artery with suture embolus method,and then reperfusion after 30-min ischemia.Rats in the sham-operated group only underwent the ligation of the common carotid artery,with no embolus plugged.After the success of the model establishment,rats in valerian ligusticum pill group were intragastrically given valerian ligusticum pill(50mg/kg)for 7days,while an equal amount of distilled water was given to rats in the sham-operated group and model group.The expression levels of Nestin,DCX and NeuN were detected in the cerebral ischemic penumbra of rats by immunofluorescent staining.Results The expression levels of Nestin were(18.67±0.58),(49.00±5.29),and(105.67±3.51)in sham-operated group,model group and valerian ligusticum pill group,respectively,with significant difference found between the valerian ligusticum pill group and the model group(P〈0.01).The expression levels of DCX were(21.67±1.53),(61.00±4.36),and(112.00±12.53)in sham-operated group,model group,and valerian ligusticum pill group,respectively.The expression levels of NeuN were(28.67±6.51),(46.00±4.36),and(323.00±10.82)in sham-operated group,model group and valerian ligusticum pill group,respectively.There were significant differences in the expression of DCX and NeuN between the valerian ligusticum pill group and model group(P〈0.01).Conclusion Valerian ligusticum pill may participate in the repair process of brain tissue injury by inducing the expression of Nestin,DCX and NeuN and promoting the neurogenesis in the cerebral ischemic penumbra.
