摘要
目的建立用于测定成骨细胞内外淫羊藿苷和朝藿定A、B、C含量的方法,并比较成骨细胞对这4种成分的吸收利用差异。方法采用UPLC法测定含量。色谱条件:Capcell C18色谱柱(250 mm×4.6 mm,5μm),乙腈-3%醋酸(24∶76)为流动相,流速为1 mL·min^(-1),检测波长为270 nm。采用外标法定量。结果 4种淫羊藿黄酮苷与培养基及细胞中的其他成分能很好分离。朝藿定A、B、C和淫羊藿苷分别在0.912~18.24 mg·L^(-1)、1.032~20.64 mg·L^(-1)、1.320~26.40 mg·L^(-1)和1.024~20.48 mg·L^(-1)内线性关系良好,平均回收率分别为97.92%、98.87%、96.36%和96.54%。给药培养24 h的成骨细胞对朝藿定A、B、C和淫羊藿苷的平均吸收率为11.94%、17.58%、37.69%和32.30%,平均转化利用率为80.49%、92.21%、87.50%和95.23%。结论建立的方法准确、灵敏,可为淫羊藿在成骨细胞内的药动学研究提供检测方法;4种黄酮苷中,成骨细胞对淫羊藿苷的吸收和利用最好。
AIM To establish a method for quantitative determination of Epimedin A, B, C and Icariin inside and outside of osteoblasts, and study the absorption and utilization of the four components in osteoblasts. METHODS The quantitative method of four Epimedii Folium flavonoid glycosides by UPLC was established. The samples were sepa- rated on a Cls column (250 mmx4.6 mm, 5 μm) with the mobile phase consisted of acetonitrile-3% acetic acid (24: 76), the flow rate was lmg·L-1, the detection wavelength was 270nm, and the external standard method was used. RESULTS Four Epimedii Folium flavonoid glycosides could be well separated from the other components of the culture medium and cells. In the range of 0.912- 18.24 mg·L-1 of Epimedin A, 1.032-20.64 mg·L-1 of Epimedin B, 1. 320 - 26.40 mg·L-1 of Epimedin C and 1. 024 - 20.48mg·L-1 of Icariin respectively, there were in good linear re- lationship, the average recovery rate was 97.92 %, 98.87 %, 96.36 % and 96.54 % respectively. The osteoblasts were treated by four Epimedii Folium flavonoid glycosides for 24 h. The average absorption rate was 11.94 % for Epimedin A, 17.58% for Epimedin B, 37.69% for Epimedin C and 32.30% for Icariin, the average utilization rate was 80.49%, 92.21%, 87.50 % and 95.23 %, respectively. CONCLUSION The method is accurate and sensitive, and can provide a reference for the studv of dru dvnamics in osteoblasts. The results suzest that icariin has showed the best absorotionand utilization for osteoblasts in four flavonoids.
