摘要
哮喘是一种常见的多发呼吸系统疾病,其患病部位在气管、支气管、肺部及胸腔。苯乙醇胺类药物是治疗哮喘的一线药物,是临床应用最广的支气管扩张剂。这类药物是一种选择性的肾上腺素受体激动剂,具有很好的疗效和广阔的市场前景。在国内外文献报道和专利中,苯乙醇胺类药物的合成方法很多。结合药物骨架的结构特点,将其归纳为5种不同的合成路线,并论述了这5种合成方法的研究进展及其优缺点。同时,对合成方法手性中心的合成和适用范围进行了总结,对药物开发提供有益帮助。
Asthma is a kind of common and frequently occurring respiratory disease,whose diseased areas is located in trachea, bronchus, lung and pleura. Phenylethanolamine-type drugs are first-line medicines, which are used as bronchodilator in clinical application. These medicines are a kind of selective adrenergic receptor agonist, which has good curative effect and broad market prospects. In the domestic and international literatures and patents,there included many synthetic methods. Coinbined with structural characteristics of drug skeletons, synthetic methods were concluded into five different synthetic routes, development of these synthetic methods were reviewed, and the advantages and disadvantages were evaluated. Furthermore, the asymmetric method of ehiral center and scope of application were discussed. By the discussion of synthetic methods,it could provide benefit to drug research and synthesis.
