参桂保心浓煎剂对心衰大鼠NT-pro BNP及Caspase-3的影响 被引量:13

Effects of ShenGui BaoXin Concentrated Decoction on NT-pro BNP and Caspase-3 of the Rats with Heart Failure

摘要 目的:探讨参桂保心浓煎剂对异丙肾上腺素(Isoproterenol,ISO)诱导左心室功能不全大鼠血清N-端脑钠肽前体(NT-pro BNP)和心肌半胱氨酸天冬氨酸蛋白酶3(Caspase-3)的影响。方法:将50只雄性Wistar大鼠随机分模型组,低、中、高剂量组和空白组,每组10只。模型组予致心力衰竭剂量(85 mg/kg)ISO后给予等容量的生理盐水,空白组不予任何干预,余各组ISO给药后予低、中、高剂量的参桂保心浓煎剂治疗,8周后采用ELISA法测NT-pro BNP含量,HE染色观察心肌组织形态学,免疫组化检测Caspase-3蛋白表达。结果:1)与模型组相比,各药物组均可降低NT-pro BNP水平(P<0.01),但仍高于空白组(P<0.01),与中剂量相比,低剂量组无明显差别(P>0.05),高剂量组显著下降(P<0.01)。2)与模型组相比,各药物组均能抑制Caspase-3表达(P<0.01),但仍高于空白组(P<0.01),其抑制作用高剂量组强于中剂量组(P<0.01),中剂量组强于低剂量组(P<0.05)。结论:参桂保心浓煎剂能降低ISO诱导的左心室功能不全大鼠NT-pro BNP水平,改善心功能,可能机制是抑制Caspase-3过度表达,阻断细胞凋亡。 Objective: To explore the effects ofShenGui BaoXin concentrated.decoction on NT-pro BNP and Caspase-3 of isoproterenol (ISO)-induced rat model with left ventricular insufficiency. Methods: Fifty rats were ran- domized into the model group, low, moderate and high doses groups and the blank group, ten rats each group. The rats in the model group were given the same amount of physiological saline after administered them ISO in the dose inducing heart failure (85 mg/kg), the rats of the blank group were unhandled, the other groups were treated with ShenGui BaoXin concentrated decoction in low, moderate and high doses after giving ISO, the contents of NT-pro BNP were detected by ELISA method in eight weeks, myocardial histomorphology was observed by HE staining, the expressions of easpase-3 were detected by immunohistochemical method. Results: 1) Compared with the model group, the level ofNT-pro BNP decreased in different medication groups (P〈0.01), but it was higher than that of the blank group obviously (P〈0.01), compared with the moderate dose group, there was no obvious difference in the low dose group (P〉0.05), it lowered in the high dose group significantly (P〈0.01). 2) Compared with the mod- el group, the expressions of Caspase-3 were inhibited in different medication groups (P〈 0.01), but they were higher than these of the blank group obviously (P〈0.01), the high dose group was stronger than the moderate dose group in the inhibiting effects (P〈0.01), the moderate dose group was stronger than the low dose group (/'〈0.05). Conclu- sion: ShenGui BaoXin concentrated decoction could decrease the level of NT-pro BNP of ISO-induced rat model with heart failure and improve heart function, its mechanism might be inhibiting overexpression of Caspase-3 and blocking cellular apoptosis.
