广西崇左市山黄皮种质资源调查及ISSR遗传多样性分析 被引量:3

摘要 【目的】分析广西崇左市山黄皮种质资源的遗传多样性,为山黄皮资源的分类、鉴定、保护和开发利用提供理论依据。【方法】调查和收集广西崇左市龙州县、宁明县、凭祥市、江州区、天等县、扶绥县和大新县7个县(市、区)的山黄皮种质资源,并进行ISSR-PCR扩增、聚类分析和主成分分析。【结果】15条多态性ISSR引物可从50份山黄皮种质资源中扩增出136条带,多态性条带比例为55.15%;各山黄皮种质间的相似系数在0.769,4).942,平均0.875。聚类分析结果显示,ISSR分子标记能将供试材料完全区分,在遗传相似系数0.864处,可将50份山黄皮种质资源聚为A、B两大组,其中A组包括来自龙州县、宁明县、凭祥市、江州区和扶绥县的40份种质,B组包括来自天等县和大新县的10份种质。主成分分析结果表明,前三主成分贡献率依次为12.11%、8.73%和7.68%,累积贡献率为28.52%;主成分分析可将50份山黄皮种质资源分为两大类,与聚类分析结果基本一致。【结论】广西崇左市山黄皮种质资源较丰富,大部分山黄皮种质亲缘关系较接近,且与地理起源有一定的相关性,NM9、TD1、DX2和JZ5等遗传差异较大的种质可作为山黄皮优良品种和单株选育及良种繁育等研究的重要材料。 [Objective]The present experiment was conducted to analyze genetic diversity of Clausena indica (Dalz.) Oliv germplasm resources in Chongzuo, Guangxi, in order to provide theory basis for its classification, identification, protection and utilization. [ Method ] C. indica germplasm resources were investigated, which were collected from Longzhou county, Ningming county, Pingxiang city, Jiangzhou region, Tiandeng county, Fusui county and Daxin county of Chongzuo city in Guangxi, the collected accessions were amplified using inter-simple sequence repeats (ISSR) molecular markers by PCR, and then the cluster analysis and principal component analysis(PCA) were conducted to detect genetic diversity and relationship. [Result]A total of 136 bands were amplified from 50 accessions of C. indica germplasms using 15 polymorphic ISSR primers, 75 of which were polymor-phic loci and with a polymorphism rate of 55.15%. The similarity coefficient between the germplasms varied from 0.769 to 0.942 ,with an average of 0.875. The cluster analysis conducted by unweighted pair-group method with arithmetic means (UPGMA) showed that ISSR markers could distinguish tested C. indica germplasms from each other. 50 accessions of C. indica germplasm were divided into 2 groups (Group A, B ) at the genetic similarity coefficient of 0.864, Group A included 40 accessions of gremplusms from Longzhou county, Ningming county, Pingxiang county, Jiangzhou region, Fusui county, and Group B included 10 accessions of germplasms from Tiandeng county and Daxin county. 4. The results of PCA showed that, the contribution rates of the first three principal components were 12.11%,8.73% and 7.68% in sequence, and the accumulated contribution rate was 28.52%. PCA also clustered 50 accessions of germplasms into 2 groups, which was similar to that of cluster analysis. [ Conclusion ] C. indi- ca germplasm resources from Chongzuo, Guangxi is abundant, and ISSR molecular markers can be used as effective tool to classify and identify C. indica germp
