家蚕脂肪酶Bmlipase-1基因在抗血液型脓病不同遗传类型材料中的表达分析及其在育种中的应用 被引量:2

摘要 为了研究家蚕脂肪酶Bmlipase-1基因表达量与家蚕抗核型多角体病毒病(又称血液型脓病)能力的关联性,以抗性品种‘KNC’和敏感品种‘HYB’及其相互交杂组配材料为试验材料,利用添毒实验与荧光定量PCR技术,检测家蚕不同遗传类型材料在不同感染条件下对血液型脓病的抵抗能力及其相应的Bmlipase-1基因表达水平。结果表明:Bmlipase-1表达水平高低与材料的抗性强弱相一致,抗性强的材料的Bmlipase-1表达量高,反之则低;抗性品种‘KNC’、‘KNC’בHYB’和‘HYB’בKNC’在添毒后的Bmlipase-1的表达量与未添毒相比有显著上调,而‘HYB’仅微量上调。Bmlipase-1的表达可以在家蚕育种过程中作为家蚕抗核型多角体病毒材料选择与鉴定的依据之一。 In order to study the relationship between expression level of Bmlipase-1 gene and resistance of silkworm to Bombyx mori nucleopolyhedrovirus(Bm NPV), a resistant variety‘KNC', a sensitive variety‘HYB'and the hybrid strains of the two varieties were selected as materials. The resistance to Bm NPV and thecorresponding expression level of Bmlipase-1 gene were detected by virus(Bm NPV) attack test and fluorescentquantitative PCR(q RT- PCR) technology. The results showed that the expression level of Bmlipase- 1 wasconsistent with the strength of resistance. The expression level was higher in silkworm varieties with strongerviral resistance, otherwise it was lower. The expression level of Bmlipase-1 had significant differences between‘KNC'and‘HYB'. After infection of Bm NPV, the expression of Bmlipase-1 in‘KNC',‘KNC'×'HYB'and‘HYB'×'KNC'was significantly higher than that of the variety with non-infection of Bm NPV. However, theexpression of Bmlipase-1 in‘HYB'was only slightly higher. It was concluded that the expression of Bmlipase-1 could be one of the basis in selecting anti-Bm NPV material in silkworm breeding.
