补肾健脾方对免疫性卵巢功能衰退小鼠性激素及卵子质量的影响 被引量:17

Effect of Bushen Jianpi Fang(补肾健脾方) on Sex Hormone and Oocyte quality in Mice with Immune Ovarian Function Decline

摘要 目的探讨补肾健脾方治疗卵巢储备功能减退的可能作用机理。方法 60只小鼠随机分为空白组、模型组、倍美力组和补肾健脾方高、中、低剂量组,每组10只。除空白组外,其余各组建立免疫性卵巢功能衰退小鼠模型。于造模后1周开始补肾健脾方低、中、高剂量组分别给予浓度为0.54、1.08、2.16 g/ml的补肾健脾方灌胃,每次0.3 ml,倍美力组给予倍美力片溶液(浓度0.002 mg/ml)0.3 ml灌胃,空白组和模型组均予生理盐水0.3 ml灌胃,连续90天,每周休息1天。测定各组小鼠血清卵泡刺激素(FSH)、黄体生成素(LH)、雌二醇(E2)水平,测量卵巢重量并进行卵巢组织HE染色,计数卵母细胞数目,电镜下观察卵巢组织形态学改变。结果与正常组比较,模型组小鼠血清FSH、LH水平明显升高,E2水平明显降低,卵巢重量降低,卵母细胞数减少(P<0.01)。与模型组比较,补肾健脾方高剂量组与倍美力组均能降低血清LH与FSH水平,升高E2水平,提高卵巢重量和卵母细胞数(P<0.01)。电镜显示,模型组小鼠的卵母细胞透明带厚度不均匀,边界不清晰,可见到髓样小体和许多微绒毛;补肾健脾方高剂量组和倍美力组小鼠的卵母细胞透明带厚度比较均匀,边界也比较清晰。结论补肾健脾方可调节卵巢储备功能下降小鼠的生殖激素水平,提高卵子数量与质量,可能是其治疗卵巢储备功能减退的作用机理之一。 Objective To investigate the possible mechanism of Bushen Jianpi Fang( 补肾健脾方) in the treatment of ovarian reserve function decline. Methods Sixty mice were randomly divided into blank group,model group,premarin group and Bushen Jianpi Fang high,middle and low dose groups,with 10 mice in each group.Except for blank group,the other groups were established into mice model with immune ovarian function decline.Experiment began one week after the modeling. Bushen Jianpi Fang high,middle and low dose groups were given0. 54 g / ml,1. 08 g / ml,or 2. 16 g / ml of Bushen Jianpi Fang for gavage respectively,0. 3 ml each time. Premarin group was given 0. 3 ml of premarin tablet solution( concentration of 0. 002 mg / ml) for gavage,while the blank group and model group were given 0. 3 ml of normal saline for gavage,respectively. The gavage lasted for 90 days,with one day off every week. Serum levels of follicle stimulating prime( FSH),luteinizing hormone( LH) and estradiol( E_2)were tested,weight of ovary was measured,and ovarian tissues were stained with HE staining to count the number of oocytes and observe the ovarian morphology change under the electron microscope. Results Compared with those in the normal group mice,serum levels of FSH and LH were significantly increased,level of E_2 was significantly decreased,the ovarian weight was decreased,and the number of oocytes was decreased in the model group( P 〈0. 01). Compared with those in the model group,serum levels of LH and FSH reduced,level of E_2,weight of ovary and the number of oocytes increased in both the Bushen Jianpi Fang high dose group and premarin group( P 〈0. 01). Electron microscopy showed that the thickness of the oocyte zona pellucida of mice is not uniform,the boundary is not clear,with the myeloid bodies and many microvilli in the model group,but the thickness of the oocyte zona pellucida was more uniform,and the boundary was clear in the Bushen Jianpi Fang high dose group and the premarin group.
