响应面法优化裂殖壶菌突变株的发酵培养基

Response Surface Method for Optimization of the Fermentation Medium of the Mutant Strain

摘要 通过单因素试验和响应面设计相结合,对裂殖壶菌(Schizochytrium limacinum)突变株产DHA的发酵培养基进行了优化。首先通过单因素试验考察发酵培养基主要成分为葡萄糖、谷氨酸钠、酵母膏和海水晶。经响应面优化发现,当发酵培养基中葡萄糖85.99 g/L、谷氨酸钠11.86 g/L、酵母膏8.16 g/L、海水晶20.16 g/L,DHA产量理论预测值可达到6.10 g/L,优化后的DHA产量比优化前提高了11.13%。在50 L发酵罐上发酵培养,DHA产量为10.32 g/L发酵液,为摇瓶培养时的1.69倍。 Optimize the fermentation medium of Schizochytrium limacinum mutant strain to produce DHA using single factor experiment associated with response surface design.First,after the single factor experiment,the main components of fermentation medium are glucose,sodium glutamate,yeast extract,and sea crystal.Through response surface optimization,it is found that when the contents of glucose,sodium glutamate,yeast extract,and sea crystal in fermentation medium are 85.99,11.86,8.16 and 20.16 g/L,respectively,the theoretical prediction of DHA production is 6.10 g/L.After optimizing,the production of DHA increases 11.13% than before.When fermentation culture in 50 L fermentation tank,the production of DHA is 10.32 g/L and 1.69 times of it in shaking flask culture.
