摘要
试验以新疆喀什地区石榴皮为研究对象,以其抑菌活性为跟踪指标,通过系统萃取、硅胶柱层析、薄层层析、生物自显影等方法对石榴皮中抑菌物质进行分离纯化及成分初探。结果表明,系统分离各萃取相中,正丁醇萃取部分和萃取后剩余部分抑菌效果较好;静态和动态试验表明:以95%乙醇-甲醇-V(甲醇)︰V(乙酸)(3︰1)-蒸馏水-V(蒸馏水)︰V(乙酸)(3︰1)作为洗脱剂可将抑菌物质逐级分离,并且95%乙醇、甲醇、V(甲醇)︰V(乙酸)(3︰1)洗脱剂所得洗脱液的抑菌活性较强;95%乙醇洗脱剂所得流分的最佳展开剂为V(正丁醇)︰V(甲醇)=3︰7;溶剂和薄层板定性试验初步判断,95%乙醇洗脱剂所得流分中可能存在的抑菌物质为:生物碱、黄酮、酚类、物质E,在该展开剂中的Rf值分别为:0.545,0.550,0.585和0.632;其中,物质E表现出非常强的抑菌活性。
The test took pomegranate peel picked in Xinjiang Kashiregion as the experimental object,mainly analyzing and studying the activity of its antibacterial substances.Antibacterial substances was isolated and purified by series of methods such as extraction system,silica gel column chromatography,thin layer chromatography,autoradiography and other methods from pomegranate peel in this study.The results showed that N-butanol and the rest of some extracted substances had significant antibacterial activity in the phase separation of the extraction system;Antibacterial substance could be separated step by step by elution such as 95% ethanol-methanol-methanol︰acetic acid(3︰1)-distilled water-distilled water︰acetic acid(3︰1),and a strong antibacterial activity was eluted from the first three elution agent in the static and dynamic tests;The N-butanol︰methanol was equal to 3︰7,the best launch agent for the 95% ethanol elution fractions;There existed such antibacterial substance as alkaloids,flavonoids,phenols,material E in 95% ethanol elution fractions and in the developing solvent shift,the ratio values were 0.545,0.550,0.585 and 0.632;Where material E showed very strong antibacterial activity.
