Distribution of Mucus in the Airway of Pigs at Different Age

摘要 呼吸道黏液对维持呼吸道健康至关重要,黏液分泌过多是慢性呼吸道炎症性疾病的重要特征。杯状细胞分泌的MUC5AC是呼吸道黏液的主要成分。为进一步了解猪呼吸道黏液的功能及分布规律,研究随机选取3头50日龄(D50)和5头200日龄(D200)健康猪,采集肺组织固定后制作石蜡切片,进行AB-PAS染色和免疫组化。结果发现:1)呼吸道黏膜上皮黏液主要呈酸性,黏膜下层腺体存在酸性、中性黏液及其混合物;2)MUC5AC主要存在于呼吸道黏膜上皮,少量存在于黏膜下层,统计发现两组猪的细支气管黏膜上皮单位面积内杯状细胞的数目差异不显著;3)随着呼吸道管径的减小,杯状细胞、呼吸道黏液及腺体含量逐渐减少至消失。 Airway mucus is essential for maintaining the health of the respiratory tract, airway mucus hypersecretion is a hallmark of chronic airway inflammatory diseases. MUC5 AC secreted by goblet cells is the main component of airway mucus. In order to understand the function and the distribution of airway mucus better, 3 heads 50-days pigs(D50) and 5 heads 200-days pigs(D200) were randomly selected in this study, the lung sample were fixated by paraffin embedding for AB-PAS staining and immunohistochemistry. The results showed that: 1) airway mucosal epithelium mainly contains acidic mucus and the submucosal glands was composed of acidic, neutral and mixed mucus; 2) MUC5 AC mainly exists in the airway mucosal epithelium and a small amount in the submucosal glands, no significant difference of the number of goblet cells which in mucosal epithelium of bronchiole was found between the two groups of pigs; 3) the number of goblet cells, airway mucus and glands become disappear with the decrease of the diameter of the respiratory tract.
