摘要
用WPM为基本培养基 ,分别附加不同的植物生长调节剂。四种处理为 0 2 2mg/l 2 ,4—D+5mg/l 2iP ;0 .2 2mg/l 2 ,4—D +0 .5mg/l TDZ ;0 .1 8mg/l NAA +0 .5mg/l TDZ ;0 .5mg/l TDZ。使用外植体为杜鹃 (Rhododendronsimsii)一栽品种′HellmutVogel′的组培苗叶愈伤组织。试验结果是在处理NAA +TDZ和TDZ上诱导出不定芽。所用外植体为来自杜鹃 (Rhododendronsimsii) 5个栽培品种 ,即′Paloma′ ,′Mme .A .Haerens′,′Lara′,′PetrickAlba′和′FrauUrsulaHerrman′和组培苗叶愈伤组织。结果是除栽培品种′Lara′外 ,其余四个栽培品种都诱导出不定芽 ,其中两个栽培品种仅在处理NAA +TDZ上形成不定芽。总体上看 ,无论是在维持愈伤组织生长方面 ,还是在诱导不定芽形成方面 ,处理NAA +TDZ都优于处理TDZ。
Using the WPM as the basic culture medium to which different plant growth regulator were added.Four treatments were 0.22mg/l 2,4-D+5mg/l 2iP;0.22mg/l 2,4-D+0.5mg/l TDZ respectively.Explants used were In Vitro leaf callus which came from one cultivar′Hellmut Vogel′ of azalea(Rhododendron simsii).The result was that adventitious buds or shoots were induced on two treatments, i.e. NAA+TDZ and TDZ.Explants tested were In Vitro leaf callus which came from five cultivars,i.e.′Paloma′,`Mme.A.Haerens',`Lara',`Petrick Alba'and `Frau Ursula Herrman',of azalea(Rhododendron simsii).The results were that,execption of cultivar`Lara',the other four cultivars were given adventitious buds or shoots,two cultivars of them were induced to give adventitious buds or shoots only on the treatment of NAA+TDZ.On the whole,the treatment NAA+TDZ was better both in maintaining callus growth and in inducing the formation of adventitious buds than the treatment TDZ.
