MiRNA-351在小鼠卵泡发育中的调控作用实验

摘要 目的探究Mi RNA-351在小鼠卵泡发育中的调控作用。方法选取60只雌性白鼠开展实验,分别于出生第5、7、11、14天和第20天处死(每次处死12只),获取卵巢后经相应处理并采取RT-PCR测定不同时刻Mi RNA-351表达情况,并分析该基因表达在小鼠卵泡发育中调控作用。结果不同卵泡发育阶段Mi RNA-315表达水平差异均有统计学意义(P<0.05);不同卵泡发育阶段颗粒细胞凋亡水平差异有统计学意义(P<0.05);和Mimics-351组增殖效能相比较,Inhibitor-351水平远远降低,两组间差异有统计学意义(P<0.05)。结论 Mi RNA-351在小鼠卵泡发育各个过程中可能均有参与,且对增殖和凋亡水平均有调控作用,值得进一步深入探究。 Objective To explore the regulatory effect of Mi RNA-351 in mouse follicular development. Methods60 female rats were selected to carry out the experiment, which executed at birth(5d, 7d, 11 d, 14 d, 20 d, and) at birth(12 rats each).The expression of Mi RNA-351 in different time was determined by RT-PCR after obtaining ovary, and the regulation of the gene expression in mouse follicle development was also analyzed. Results The differences of Mi RNA-315 at different stages of follicular development were statistically significant(P〈0.05).There were significant differences in the levels of apoptosis of granulosa cells in different stages of follicular development(P〈0.05).The level of proliferation potency in the Inhibitor-351 group was far lower than that in the Mimics-351 group,and the difference was statistically significant between the two groups(P〈0.05). Conclusion Mi RNA-351 may be involved in various processes of mouse follicular development, and the level of proliferation and apoptosis are regulated, which is worth further exploration.
