利用核定位信号提高非病毒基因载体转化效率的研究进展

Research Progress of Using Nuclear Localization Signal to Improve the Efficiency of Non-Virus Gene Transformation

摘要 关于非病毒基因载体运用一直是基因工程研究关注的焦点,外源性基因穿过细胞膜屏障的方法较为成熟,而通过细胞核屏障的效率较低,通常需要借助核定位信号来完成。目前采用的方法各不相同,可导致核定位信号对转化效率的影响因素众多,如核定位信号与外源性DNA的连接方式、核定位信号的数量、类型、修饰,外源性阳离子聚合物、pH值等。因此,如何利用核定位信号中有效提高非病毒基因的转化效率值得研究探讨,本文就此作一综述。 The utilization of non-virus gene carrier has always been the focus during genettc engineering research,the methods of exogenous genes across the cytomembrane barrier were relatively ma- ture,but the efficiency was low when through the nucleus barrier, which always need approving by the means of nuclear localization signal(NLS). There were many impact factors that lead to a large difference of the transformation efficiency during the different methods of all experimental groups, for example, the connected methods of NLS and exogenous DNA,the amount,type,modi- fication of NLS,exogenous cationic polymer,pH value and so on. Therefore, the methods which were made full use of the NLS to improve the efficiency of non-virus gene transformation were worthy of study, this paper summarized these as following.
