舍曲林对缺血性脑卒中后抑郁模型大鼠海马TrkB和CREB表达的影响 被引量:8

摘要 目的观察舍曲林对缺血性脑卒中后抑郁模型大鼠行为学的影响,以及该模型大鼠海马酪氨酸激酶受体B(Trk B)和c AMP反应原件连接蛋白(CREB)表达的影响。方法将40只成年雄性SD大鼠随机分为假手术组、抑郁组、PSD组和舍曲林预防组,每组10只。采用改良双侧颈总动脉结扎术制备全脑缺血大鼠模型,慢性不可预见性温和刺激和孤养法制备抑郁大鼠模型,将以上方法相结合制备PSD大鼠模型。舍曲林预防组予舍曲林水溶液[1 mg/(100g·d)]对制备的PSD大鼠每天灌胃1次。观察各组大鼠自发性行为学改变,采用蔗糖水消耗试验、旷场试验、体重变化评定大鼠的行为学变化;21 d后采用免疫组织化学法分析大鼠海马Trk B、CREB蛋白的表达。结果应激开始后第7 d、第14 d、第21 d测大鼠体重、蔗糖水饮用量、旷场试验水平运动距离,PSD组较假手术组差异有统计学意义(P<0.01),预防组较PSD组差异有统计学意义(P<0.01)。免疫组化结果显示,PSD组大鼠模型海马各区Trk B、CREB的平均光密度值均明显低于假手术组(P<0.01,P<0.05),舍曲林预防组大鼠模型海马各区Trk B、CREB的平均光密度值明显高于PSD组(P<0.01)。结论舍曲林可有效改善PSD大鼠的抑郁状况,Trk B、CREB参与此过程并起着重要作用。 Objective To observe the effects of sertraline on the behavior in ischemic post-stroke depression rats,and the expression of Trk B and CREB in hippocampus.Methods 40 adult male Sprague-Dawley rats were randomly divided into control group,simple depression group,post-stroke depression(PSD) group and sertraline prevention group(n = 10each).The complete cerebral ischemic rat models were established with reformed occluding the bilateral carotid artery method;applying chronic unpredictable mild stimulation and isolation feed were used to make the rat depression model:the combination of the above methods were taken to induce rat PSD model;sertraline prevention models of rats were induced by subcutaneous gavaged with 1 mg/100(g.d) sertraline.The changes in spontaneous behavior of rats were observed,the changes of behavior in rats were evaluated by sucrose water consumption,open field test and weight changes,and the changes of tyrosine kinase receptor B(Trk B),c AMP response element binding protein(CREB) of hippocampus were measured by immunohistochemistry after 21 days.Results The rat body weight,sucrose water consumption,open field test horizontal movement distance on the 7th,14 th,21th were measured after CUMS,the PSD group compared with the control group,the differences had statistical significance(P〈0.01);the sertraline prevention group compared with the PSD group,the differences had statistical significance(P〈0.01).The results of immunohistochemistry showed that the average optical density value of both the Trk B and CREB in the rat hippocampus each area of the PSD group decreased significantly compared with control group(P〈0.01 or P〈0.05),and those of sertraline prevention group increased clearly compared with the PSD group(P〈0.01).Conclusion Sertraline may effectively improve PSD rat depression behavior,Trk B and CREB are involved in this process,and play an important role.
