联合补充维生素D胶原肽和钙对大鼠骨骼发育影响的实验研究 被引量:1

Study on the impact of combined supplementation of vitamin D,collagen peptide and calcium on bone development in rats

摘要 目的探讨补充维生素D、胶原肽和钙对大鼠骨骼发育的影响。方法健康雄性初断乳SD大鼠按体质量随机分为5组,其中1组为低钙对照组,实验期间给予低钙饲料喂养,其余4组在低钙饲料的基础上,分别给予含不同剂量的维生素D、胶原肽和钙的饲料喂养,12周后测定大鼠骨骼生长发育相关指标、血清骨代谢生化指标、钙表观吸收率,并对骨组织切片进行病理学观察。结果各受试物剂量组大鼠股骨中点骨密度为(1 889±0.011 0)g/cm2、(2 188±0.020 4)g/cm2、(1 908±0.011 4)g/cm2、(1 919±0.009 7)g/cm2,均显著高于低钙对照组,最大载荷分别为(96.95±11.85)、(122.81±13.67)、(97.48±8.94)、(100.45±19.51)N,显著高于低钙对照组,维生素D和碳酸钙联合干预组各指标优于其他各组;碳酸钙、维生素D和胶原肽三者联合干预组血清I型前胶原氨基端前肽(PINP)水平显著低于对照组;各组钙表观吸收率分别为(93.87±1.70)%、(93.72±1.20)%、(92.47±1.33)%、(90.45±2.81)%,其中2、3组比对照组(90.86±4.19)%高(P<0.05)。结论单独补充碳酸钙,或碳酸钙与维生素D和/或胶原肽联合补充均能够促进成长期雄性大鼠骨的发育,但碳酸钙、维生素D和胶原肽三者联合的干预效果未达预期,其机制有待进一步深入探讨。 Objective To observe the effect of combined supplementation of vitamin D,collagen peptide and calcium on bone development in rats. Methods Early weaned male SD rats were randomly divided into five groups by weight,including one low-calcium control group in which rats were only given low calcium feeding during the experiment,and rats in the remaining four groups were given different doses of vitamin D and collagen peptide besides the same low-calcium feeding.The rats′bone growth and development related indexes,biochemical indexes of serum bone metabolism and apparent calcium absorption rate were measured after 12 weeks,and pathological observation was conducted on the bone tissue sections. Results The bone mineral densities(BMD)of femur distal end of rats in 4experimental groups were(1 889±0.011 0),(2 188±0.020 4),(1 908±0.011 4)and(1 919±0.009 7)g/cm2,which were significantly higher than that in low calcium control group,and the ultimate load of the femur were(96.95±11.85),(122.81±13.67),(97.48±8.94)and(100.45±19.51)N,which were also significantly higher than that in low calcium control group.Combined supplementation of vitamin D with calcium had the greatest effect on bone development,while PINP level was much lower in rats supplemented with calcium,vitamin D and collagen peptide compared to control group.The apparent calcium absorption rates in each intervention groups were(93.87±1.70)%,(93.72±1.20)%,(92.47±1.33)% and(90.45±2.81)%,and the rates in group 2and group 3were higher than that of(90.86±4.19)%in control group(P〈0.05). Conclusions Suplementation with calcium alone or combined with vitamin D and/or with collagen peptide can improve bone growth of male rats,but the effect of three combined supplements is unable to meet the expectation with unknown mechanism.
