摘要
天然反义转录物(natural antisense transcripts,NATs)被定义为在自然状态下生物体内产生的内源性RNA,其通常是通过与靶分子进行序列互补而形成天然的正义-反义转录物配对的双链RNA。NATs普遍存在于原核和真核生物中,可以在转录水平、转录后水平、翻译水平等多个层次经由不同机制调节基因的表达。此外,NATs也与恶性肿瘤的发生发展密切相关,一些NATs在正常组织与神经胶质瘤组织中的表达存在显著差异。NATs可能成为神经胶质瘤诊断、预后以及治疗效果的一种新的标志物,同时NATs也是潜在的新的胶质瘤基因治疗的靶点。本文对NATs的分类及其在神经胶质瘤中的功能及可能的机制做具体阐述。
Natural antisense transcripts (NATs) are reverse-complementary at least in part to the sequences of other endogenous sense transcripts and regulate the expression of their target genes. NATs, considered until recently as transcriptional noise, are a very common phenomenon in human and eukaryotic transcriptomes. Otherwise, they play an indisponsible functional role in organ formation, cell differentiation, diseases and pathology. NATs have been shown to regulate nearly every level of gene regulation: pre-transcriptional, transcriptional and post-transcriptional, through DNA-RNA, RNA- RNA or pretein-RNA interactions. Glioma is a kind of brain tumor with high incidence and lethality. Recently, it had been reported the profile of NATs in glioma and indicate many NATs which were dysregulated. The roles of NATs in gliomagenesis were primarily characterized and we discovered some significant findings about maker molecules. In this review, we summarized the research on the function of oncogenic NATs in glioma and made some detail for mechanism.
