摘要
目的基于血管内皮生长因子(VEGF)相关信号通路探讨白英总碱对A549细胞的凋亡与周期的影响。方法设立对照组和白英总碱低、中、高剂量组(50,100,200 mg·L^(-1)),通过流式细胞术检测细胞凋亡及周期分布情况,Western blot法检测PI3K、Akt、Ras、MAP2、VEGF蛋白的表达。结果流式细胞术检测白英总碱将该细胞周期阻滞于G_2期,呈现剂量依赖性的诱导A549细胞的凋亡,尤其是早期凋亡细胞。Western blot结果显示,白英总碱均可下调PI3K、Akt、Ras、MAP2、VEGF蛋白的表达,中、高剂量组各蛋白的相对表达量与对照组比较,差异均有统计学意义(P<0.01)。结论白英总碱可通过调控VEGF相关信号通路诱导A549细胞的凋亡,阻滞细胞周期于G_2期。
Objective To investigate the effect of total alkaloids from Solanum lyratum on the apoptosis and cell cycle of A549 cells based on vascular endothelial growth factor(VEGF)-related signal pathway. Methods Control group and three experimental groups were set up,cell cycle and apoptosis were tested by flow cytometry,and protein levels of PI3 K,Akt,Ras,MAP2,and VEGF were examined by Western blot method. Results The results of flow cytometry showed that total alkaloids from Solanum lyratum arrested the cell cycle at G_2 phase,and induced A549 cells apoptosis in a dose-dependent manner,especially for the early apoptotic cells. The results of Western blotting showed that total alkaloids from Solanum lyratum down-regulated the protein levels of PI3 K,Akt,Ras,MAP2,and VEGF,and the relative protein expression quantity in middle- and high-dose groups obviously differed from that of the control group(P0.01). Conclusion Total alkaloids from Solanum lyratum could induce A549 cells apoptosis and arrest cell cycle at G_2/M phase through VEGF-related signal pathway.
