摘要
目的观察肝纤维化模型中小鼠肝脏的病理改变以及软肝冲剂对小鼠肝纤维化模型的治疗作用。方法将60只雄性SPF级C57B/L小鼠随机分为空白对照组、模型组、护肝片对照组、软肝冲剂低剂量组和软肝冲剂高剂量组,每组12只。空白对照组给予100%花生油1 mL/kg腹腔注射,每周2次,共10周;模型组给予含20% CCl_4的花生油1 mL/kg腹腔注射,每周2次,共10周;护肝片对照组给予含20% CCl_4的花生油1 mL/kg腹腔注射,每周2次,共10周,并从第3周起给予护肝片0.1 mg/(kg·d)灌胃至第10周;软肝冲剂低剂量组给予含20% CCl_4的花生油1 mL/kg腹腔注射,每周2次,共10周,并从第3周起给予软肝冲剂1 g/(kg·d)灌胃至第10周;软肝冲剂高剂量组给药剂量为5 g/(kg·d),造模及给药方法与低剂量组相同。在第10周通过摘除眼球取血,检测小鼠血清中的透明质酸(HA)、层黏连蛋白(LN)、三型前胶原(PCⅢ)、四型胶原(Ⅳ-C)、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST);检测肝脏组织中超氧化物歧化酶(SOD)和丙二醛(MDA)的含量;计算肝脏指数;观察小鼠肝脏的HE染色和MASSON染色病理组织学改变。结果模型组与空白对照组比较,小鼠血清中HA、LN、PCⅢ、Ⅳ-C、ALT和AST水平显著升高(P<0.05),肝脏组织中SOD显著降低(P<0.05),MDA显著升高(P<0.05),肝脏指数增加(P<0.05),病理切片显示肝细胞坏死并出现脂肪空泡、纤维组织增生和炎细胞浸润,MASSON染色显示肝组织内出现明显脂肪空泡和纤维组织增生;软肝冲剂组与模型组比较,小鼠血清中HA、LN、PCⅢ、Ⅳ-C、ALT和AST的水平显著降低(P<0.05),肝脏组织中SOD显著升高(P<0.05),MDA水平显著降低(P<0.05),肝脏指数降低(P<0.05),病理检查显示肝脏组织中无炎性浸润、脂肪空泡和纤维组织沉积,MASSON染色显示肝组织内出现明显脂肪空泡和纤维组织增生。结论 CCl_4腹腔注射可以形成小鼠肝硬化模型,病理改变以脂�
Objective To observe the pathological changes of liver fibrosis model in mice and the therapeutic effect of Ruangan granule (RGG) on liver fibrosis model in mice. Methods Totally 60 male SPF class C57B/L mice were randomly divided into five groups, including control group, model group, colchicines group, RGG low group and RGG high group, 12 mice in each group. Mice in control group were injected 100% peanut oil, 1 mL/kg,twice a week in totally 10 weeks;mice in model group were injected peanut oil containing 20% carbon tetrachloride, 1 mL/kg, twice a week in 10 weeks; mice in liver-aid tablets group were injected peanut oil containing 20% carbon tetrachloride, 1 mL/kg ,twice a week in 10 weeks, and colchicines was used from the third week until the tenth week, 0. 1 mg/ (kg. d) ; mice in RGG low group were injected peanut oil containing 20% carbon tetrachloride, 1 mL/kg, twice a week for 10 weeks,and RGG was used from the third week until the tenth week,1 g/(kg-d) ;mice in RGG high group were treated in the same way as RGG low group, but RGG was fed in 5 g/( kg- d). At the tenth week, blood samples were collected from the mice by removing eyes and the content of hyaluronic acid ( HA ) , layer adhesion protein collagen type (LN) , three former 111 ( PC III ) , collagen- 1V ( 1V-C ) , alanine aminotransferase (ALT) and aspartate aminotrans- ferase (AST) in serum were tested. The content of super oxide dismutase (SOD) and malondialdehyde (MDA) in liv- er tissue was tested;liver index was calculated;pathological and histological changes of mice liver were observed by HE and MASSON staining. Results The levels of HA, LN, PC Ⅲ,Ⅳ-C, ALT and AST in serum of mice in model group were significantly increased ( P 〈 0. 05 ) compared with those in control group ; the content of SOD in liver tissue was significantly lower ( P 〈 0. 05 ) ; the content of MDA in liver tissue was significantly higher ( P 〈 0. 05 ) ; liver index was significantly higher ( P 〈 0.
